瑞舒伐他汀对急性脑缺血再灌注大鼠脑含水量及海马区MG的影响 被引量:3

Effect of rosuvastatin on microglia and cerebral water content in rats following acute cerebral ischemia-reperfusion

摘要 目的通过观察瑞舒伐他汀对急性脑缺血再灌注损伤大鼠脑含水量及海马区小胶质细胞(microglia,MG)影响,探讨其脑保护作用机制。方法将96只SD大鼠,随机分为4组:正常大鼠为A组,假模型组为B组,模型组为C组,干预组为D组。应用改良的Zea Longa线栓法制作大鼠大脑中动脉缺血再灌注(MCAO)模型,应用免疫组化法观测大鼠海马区MG改变,采用干湿重法观测脑含水量变化。利用HE染色观察海马区神经元的变化。结果 A、B组不同时间点脑含水量、MG表达无明显改变。C、D组大鼠的脑含水量、MG的表达在24 h开始增高,72 h时达到峰值,7 d开始下降,14 d明显下降,与A、B组相比较,差异明显(P<0.05)。D组与C组脑含水量、MG表达相比,24 h及72 h差异不明显(P>0.05),7 d及14 d,D组的组脑含水量、MG表达较C组下降明显,差异具有统计学意义(P<0.05)。结论瑞舒伐他汀能明显降低急性脑缺血再灌注大鼠脑水肿及MG的表达,抑制神经元过度凋亡,具有脑保护作用。 Objective To investigate the effect of rosuvastatin on cerebral water content and microglia in hippocampus of rats with acute cerebral ischemia reperfusion injury and the possible mechanism.Methods 72 SD rats were randomly divided into 4 groups:the normal rats were A group,the sham model group was B group,the model group was C group,the intervention group was D group.Rat model of middle cerebral artery ischemia reperfusion(MCAO)was made by the improved Zea Longa suture technique.MG changes in hippocampus of rats were observed by immunohistochemistry.The changes of cerebral water content were observed by wet and dry weight method.HE staining was used to observe the changes of neurons in hippocampus.Results The brain water content and the expression of MG in A and B groups were not changed at different time points.The brain water content and the expression of MG in C and D group rats began to increase at 24 h,reached peak at 72 h,7 d began to decrease,and 14 d decreased significantly,compared with A and B group,the difference was obvious.Compared with group C and group 72 h,the brain water content and the expression of MG in D group were not significantly different from those in 24 h group.The brain water content and MG expression in group 7 d group 14 d and group D were significantly lower than those in group C,andthe difference was statistically significant.Conclusion Rosuvastatin can protect brain by reducing the brain edema and expression of MG and by inhibiting the excessive apoptosis of neurons after acute erebral ischemia reperfusion in rats.
