摘要
为控制外植体褐化及促进外植体的萌芽和愈伤组织的形成,以淡水沙梨新梢为材料,以MS(Murashige and Skoog)培养基为基本培养基,添加活性炭或植物生长调节剂培养顶芽、腋芽和节间茎段.结果表明,在培养基中添加2 g/L活性炭的淡水沙梨褐化率为6.9%,而未添加活性炭的淡水沙梨褐化率为11.1%,新梢当天接种褐化率为13.3%,第二天接种的褐化率为46.7%,第四天接种的褐化率为66.7%.新梢顶芽的出芽率为48.5%,愈伤组织形成率为9.0%,腋芽的出芽率为30.3%,愈伤组织形成率为51.5%,节间无芽茎段的出芽率为0,愈伤组织形成率为45.2%.培养基中添加活性炭能降低褐化率,离体时间对外植体培养的褐化有很大影响,新梢离体时间越长,接种后褐化率越高,带顶芽的外植体比带腋芽的外植体出芽率更高.
In order to control browning, promote sprouting and the formation of callus of explant, selecting Danshui pear as material,and taking MS(Murashige and Skoog)as the basic medium, activated carbon and plant growth regulators have been added to cultivate terminal buds, axillary buds and internodal stems without buds. It has been revealed that the browning rate of fresh material is 7.2% after adding 2 g/L activated carbon tothe medium. The browning rate is 13.3% when the picked branches have been inoculated at the same day asthey were picked. And the browning rate is 46.7% and 66.7%. There is a marked difference of browning amongthat very day, the second day and the fourth day. The germination rate for terminal buds is 48.5% while formationrate for callus is 9.0% .The germination rate for axillary buds is 30.3% while formation rate for callus is51.5%. And formation rate for callus of internodal stems without buds is 45.2%. That is to say, germination rate of terminal buds is higher than that of axillary buds. The browning rate for explant is closely related to inoculative time. Low degree of browning would be achieved while inoculated at that very day to culture in vitro.
