摘要
用XhoI内切酶切出在puc18中克隆的硅藻质粒pcf_1(4.3kb),用T_4 DNA连接酶自身连接环化,再以BclI酶切后插入BamHl酶切的pSV-CAT质粒(5kb),得到了插入方向不同的2种杂种质粒(DRP3和DRP4)。改建后的杂种质粒是一种可用於硅藻和其它真核细胞遗传工程研究用的自主复制型质粒。
Diatom plasmid pcfl (4.3kb) had been inserted in Vector PSV-CAT (5kb) with partial complementary of the Bell and BamHI sticky ends. All the DNA fragments needed for recombination are recovered from agarose gel afer electrophoresis. To identify the orientation of inserted pefl in either of two directions, the hybrid plasmids were preparated from transformants, digested by HindⅢ. The electrophoresis graph (3.4) proved that both orientation of the inserted pcfl were present on the hybind plasmids. This kind of hybvid is now called Diatom Autonomously replicating vector useful as a tool for developing algae and plant genetic engineering.
