摘要
目的:研究外用芍药苷(PF)对咪喹莫特银屑病小鼠模型的治疗作用。方法:BALB/c小鼠12只随机分为空白、IMQ造模、治疗低剂量组及治疗高剂量组4组,利用咪喹莫特(IMQ)构建银屑病小鼠模型,治疗组在建模过程中同时给小鼠耳部皮肤连续外用涂抹不同浓度的PF。连续7d后分别与空白组、IMQ单纯造模组及比较,观察模型小鼠的银屑病表型变化及HE染色下皮肤组织病理改变。结果:7d后与空白组及IMQ组相比较,外用涂抹PF后小鼠红斑鳞屑明显减轻,表皮厚度明显减轻,HE显示真皮浸润淋巴细胞数量明显减少,并且PF的治疗作用随剂量的增加而增强,提示外用PF能够抑制银屑病炎症反应从而缓解银屑病表型。结论:外用PF具有治疗银屑病的作用,PF可作为银屑病靶向治疗药物进一步开发及临床应用。
Objective: To illustrate the effects of paeoniflorin as topical therapy on psoriasis-like mouse model. Methods: BALB/c mice were randomly assigned to four groups including control group, IMQ-induced group, IMQ accompanied with low-dose paeoniflorin treatment, and IMQ accompanied with high-dose paeoniflorin treatment. Shaved mouse dorsal skin of three groups was treated daily with Aldara cream and one control group with Vaseline for seven days. Two experimental groups of mice also received topical treatment with paeoniflorin. Mice were sacrificed on the seventh day and skin samples were collected for subsequent phenotype analysis and HE staining. Results: The results showed that the erythema scales and the epidermis thickness of ears were significantly reduced after topical treatment of paeoniflorin in IMQ-induced mice. HE staining also showed a significant decrease in the number of dermal infiltrating lymphocytes, and the paeoniflorin treatment exhibited a dose-dependent therapeutic effect, suggesting that topical paeoniflorin could inhibit the inflammatory responses of psoriasis and alleviate the psoriasis symptoms.Conclusion: Topical paeoniflorin alleviates the psoriasis-like inflammation in mouse model, which indicates that Paeoniflorin can be further developed and clinically applied as a targeted therapy for psoriasis.
