Optimization of extraction process of total flavonoids from Salvia prattii Hemsl.by Box-Behnken response surface methodology and evaluation of antioxidant activity

摘要 目的利用单因素实验和响应面法优选康定鼠尾草中总黄酮的提取工艺。方法采用标准曲线法测定总黄酮的含量,以提取量为参考指标,选择单因素实验筛选出乙醇体积分数、液料比和超声时间3个主要因素,通过Box-Behnken设计方案,建立总黄酮提取得率的二次回归方程,得到优化组合条件。结果最佳超声提取工艺条件为:乙醇体积分数70%,液料比:10∶1,超声时间:40min,总黄酮提取得率为6.24mg·g^(-1)。结论优选得到的康定鼠尾草中总黄酮的提取工艺合理、可行,质量稳定,有较强的抗氧化活性。 Objective The extraction technology of total flavonoids in Salvia prattii Hemsl.was optimized by single factor tests and response surface methodology. Methods The content of total flavonoids was determined by standard curve and the extraction quantity was regarded as the investigated index.In order to obtain the maximum extraction yield of total flavonoids,3 main factors including ethanol concentration,ratio of water to raw material and extraction times were selected with the Box-Behnken design and a quadric regression equation for extraction yield of total flavonoids was established,the optimum combination condition was obtained. Results The optimum extracting conditions were ethanol volume fraction 70%,liquid to material ratio 10∶1and extract time 40 min.The extraction yield of total flavonoids was 6.24 mg·g^-1. Conclusion The optimum technology is stable and feasible for the extraction of total flavonoids in Salvia prattii Hemsl.,and the total flavonoids extract showed strong antioxidant activity.
