HPLC法测定不同产地刺五加中原儿茶酸、紫丁香苷、绿原酸、刺五加苷E和异嗪皮啶 被引量:10

Determination of protocatechuic acid, syringing, chlorogenic acid, eleutheroside E, and isofraxidin in Acanthopanax senticosus from various of habitats by RP-HPLC

摘要 目的建立HPLC法测定不同产地刺五加中原儿茶酸、紫丁香苷、绿原酸、刺五加苷E和异嗪皮啶。方法采用Welch Materials C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈–0.1%磷酸水,梯度洗脱;体积流量为1.0 m L/min;柱温30℃;检测波长214 nm;进样体积10μL。测定结果采用主成分分析法进行分析。结果 5种成分在测定质量浓度范围内线性关系良好,r均大于0.999 9;平均回收率为99.66%~101.25%,RSD值为1.92%~3.03%;不同产地的11批刺五加中原儿茶酸、紫丁香苷、绿原酸、刺五加苷E和异嗪皮啶的质量分数范围分别为0.004%~0.016%、0.080%~0.152%、0.372%~0.806%、0.057%~0.103%、0.002%~0.010%。主分成分析结果显示,黑龙江产刺五加药材质量较佳。结论该方法快速、简便、准确,多成分测定的质量评价模式可用于刺五加药材的质量控制。 Objective To establish an HPLC method for simultaneous determination of protocatechuic acid, syringing, chlorogenic acid, eleutheroside E and isofraxidin in Acanthopanax senticosus(Rupr. et Maxim.) Harms from different habitats. Methods Separation was carried out on Welch Materials C_(18) column(250 mm × 4.6 μm, 5 μm). The mobile phase consisted of acetonitrile-0.1% phosphoric acid water with gradient elution. The detection wavelengths were set at 214 nm. The flow rate was 1.0 m L/min, temperature of column was set at 30 ℃, and volume of injection was 10 μL. The result was analyzed by PCA. Results The linear relations of five active components in the range of mass concentration was good with perfect precision with r all more than 0.999 9. The average recovery rates were 99.66% — 101.25%, and RSD was 1.92% — 3.03%. The contents in 11 batch of the A. senticosus from different habitats were calculated, and the results were as following: protocatechuic acid was about 0.004% — 0.016%, syringing was 0.080% — 0.152%, chlorogenic acid was 0.372% — 0.806%, eleutheroside E was 0.057% — 0.103%, and isofraxidin was 0.002% — 0.010%. Conclusion The method is rapid, convenient, and with precision and accuracy, and the multi-component determination method is suitable for the quality control of A. senticosus.
