Roux-en-y胃旁路术对2型糖尿病大鼠血清Angptl 2及B2R的影响 被引量:1

Influence of Roux-en-y gastric bypass on serum Angptl 2 and B2R in type 2 diabetic rats

摘要 目的观察Roux-en-y胃旁路术(RYGB)对2型糖尿病(T2DM)大鼠缓激肽受体2(B2R)和血管生成样蛋白2(Angptl 2)的影响。方法将60只SD大鼠随机分为糖尿病组40只和正常对照组(NC组)20只。糖尿病组大鼠给予高脂高糖饮食4周后,经腹腔注射链脲佐菌素(STZ)建立模型。将成模大鼠再随机分为实验组(RYGB组)15只(手术后死亡2只)和假手术组(DM组)15只。RYGB组大鼠行RYGB手术,DM组和NC组大鼠接受原位手术。三组大鼠分别于术前和术后4周测定三组大鼠的体重、FBG、FINS、ISI、Angptl 2及B2R水平。结果 RYGB组与DM组大鼠术前体重、FBG、FINS、ISI、ANGPTL2、B2R水平无明显差异(P>0.05)。术后4周RYGB组大鼠较术前体重、FBG、FINS均明显下降(P<0.05),ISI、ANGPTL2及B2R上升(P<0.05)。DM组、NC组大鼠术后4周的体重、FBG、FINS、ISI、ANGPTL2、B2R水平与术前比较,差异无统计学意义(P>0.05)。结论胃转流术可能是通过降低T2DM大鼠B2R和Angptl2的水平,进而达到减重、降低血糖的效果。 Objective To research the influence of Roux-en-y gastric bypass surgery on bradykinin receptor 2( B2 R) and angiogenesis like protein-2( Angptl 2) in T2 DM rats. Methods 60 healthy Sprague Dawley( SD) rats were randomly divided into diabetic mellitus group( n = 40) and normal control group( n =20). Then rats of diabetic group were fed with high calorie and high sugar diet for 4 weeks,following by one dose of streptozotocin( STZ) via intraperitioneal injection. The model rats were randomly divided into the experimental group( RYGB group,n = 15)( 2 rats died after operation) and the diabetic mellitus sham operation group( DM group,n = 15). The rats of RYGB group were undertaken with RYGB,and the rats of DM group and NC group were undertaken sham operation. The level of weight,FBG,FINS,ISI,Angptl 2 and B2 R were detected before surgery and 4 weeks after surgery.Results Before the surgery,RYGB group compared with DM group,the level of weight,FBG,FINS,ISI,ANGPTL2 and B2 R were not statistically significant( P〉0.05). 4 weeks after the surgery in RYGB group,the level of weight,FBG and FINS were decreased( P〈0. 05),and ISI,ANGPTL2 and B2 R were gradually increased( P〈0. 05).However,in DM and NC group,there was no significant difference in the level of weight,FBG,FINS,ISI,ANGPTL2 and B2 R before and 4 weeks after surgery( P〉0.05).Conclusions Roux-en-y gastric bypass surgery may reduce weight and blood glucose by decreasing the level of B2 R and Angptl2 in type 2 diabetic rats.
