Tissue Culture and Rapid Propagation of Cinnamomun burmannii

摘要 为了提高梅片树培育效率,使育苗过程不再受制于季节环境等因素。本研究探索了梅片树组培快繁的过程。通过实验发现,将带有腋芽的梅片树茎段外植体经消毒灭菌后,接种在DCR+6-BA(2 mg/L)+NAA(0.05 mg/L)+蔗糖(30 g/L)的培养基中,在温度(25±2)℃,光照强度1 500 Lx,光照时间16 h/d(6:00~22:00)的条件下培养一个月,诱导腋芽的发生,芽的诱导率达到89.83%,且平均芽长在1.5 cm以上,叶片健康舒展。将诱导出的腋芽接种于DCR+6-BA(4 mg/L)+NAA(0.05 mg/L)+蔗糖(30 g/L)的增殖培养基中,在同样条件下培养40 d,芽增殖倍数达到1.90,且所有芽均明显发育伸长,比增殖前更加健壮。将健壮枝芽接种于DCR+IBA(1.5 mg/L)+蔗糖(30 g/L)的生根培养基中,枝芽21 d即可观察到生根,30 d后生根率达到80.95%。随着根的延伸并形成根系,枝芽逐渐生长,形成无菌苗,60 d苗高度达到5 cm左右。本研究通过一种新途径实现了梅片树组织培养的再生繁殖,显著提高了梅片树的芽诱导率、繁殖系数和生根率,为梅片树的组培快繁产业化生产提供理论依据。 The study aimed to improve the cultivation efficiency of Cinnamomun burmannii trees, so that the seedling process is no longer limited by environmental factors such as seasons. The process of tissue and rapid propagation of Cinnamomun burmannii were explored in this study. Stem segment explants with axillary buds of Cinnamomun burmannii were inoculated in the medium of DCR with 6-BA(2 mg/L), NAA(0.05 mg/L) and sucrose(30 g/L) after sterilization and were cultivated one month under the conditions of(25±2)℃, 1 500 Lx illumination intensity and 16 h/d(6:00-22:00) to induce the formation of axillary bud. The result showed that the induction rate of buds was 89.83% and the average bud length was over 1.5 cm with lead stretched healthily. While when the induced axillary bud was inoculated in the medium of DCR with 6-BA(2 mg/L), NAA(0.05 mg/L) and sucrose(30 g/L) and cultivated for 40 d under the same conditions, the bud proliferation reached at 1.90 and all buds were significantly elongated which was more robust than before proliferation. When the robust shoots were inoculated in the rooting medium of DCR with IBA(1.5 mg/L) and sucrose(30 g/L), root could be observed after 21 d, and the rootage rate reached 80.95% after 30 d. As the roots extended and roots formed, buds gradually grew up to be aseptic seedlings and the height reached at 5 cm in 60 d. A new approach was adopted to regenerate the tissue culture of Cinnamomun burmannii in this study, and the bud induction rate, propagation coefficient and rooting rate of Cinnamomun burmannii were significantly increased, which might provide a theoretical foundation for the industrialized production of tissue culture and rapid propagation of Cinnamomun burmannii.
