摘要
目的:观察高脂血症能否直接诱导兔主动脉瓣膜发生退行性的改变,以及阿托伐他汀对瓣膜病变进程的影响。方法:选择健康新西兰纯种雄性,白兔,45只,随机平均分配为对照组(N组)、高脂组(HC组)和高脂加阿托伐他汀干预组(AI组)。并分别于实验开始4、8、12周后,取静脉血测定血浆脂质及hs-CRP水平,取主动脉瓣膜标本行组织学检测,经HE和免疫组织化学染色,观察钙化性主动脉瓣组织学改变及各炎性指标的变化,分析比较各组间是否存在差异。结果:第8、12周HC组和AI组血浆中TC、TG、LDL-C及高敏C反应蛋白(hs-CRP)的浓度较第4周及N组均升高,同期AI组的各项血脂水平低于HC组,差异有统计学意义(P<0.05)。与正常主动脉瓣比较,HC组光镜下组织学改变与钙化性主动脉瓣疾病早期病理改变相似,AI组较HC组病变减轻;免疫组织化学染色示:因子TNF-α、IL-6在HC组中表达明显,在AI组中的表达较HC组显著下降,N组中则无表达。结论:高脂饮食能够诱导兔主动脉瓣膜发生早期钙化并存在慢性炎症,阿托伐他汀通过降血脂及抗炎作用,能够抑制钙化性主动脉瓣疾病病变发展进程。
Objective: To observe whether the hyperlipemia can directly induce aortic valve calcific aortic valve disease( CAVD) pathological changes of experimental rabbits model and effects of atorvastatin on inflammatory factor of mitral aortic valve disease. Methods: 45 New Zealand white rabbits were randomly divided into normal control group( N group),high-fat diet group( HC group) and high-fat diet plus atorvastatin group( group AI). The level of plasma lipid of rabbits were respectively tested at beginning of 4 th,8 th and 12 th weeks. These data were compared between groups to observe whether there is significant statistical difference.Meanwhile the specimens of aortic valve were operated for histological examination. Results: The plasma glycerol TC,TG,LDL-C,C reactive protein( CRP) concentration of HC and AI group at the 8 th and 12 th weeks were increase compared to N group and the end of 4 th week. At the same period the serum lipid level of AI group was lower than the HC group. Statistical analysis showed significant difference( P〈0. 05). Compared with the HC group,the immunohistochemistry result showed that TNF-α and IL-6 in group AI was reduced,and there is no express in Group N. Conclusion: High fat diet can induce the rabbit aortic valve to pathological changes,which is similar to the early CAVD pathology. And there are inflammatory changes in the samples.Atorvastatin can reduce the expression of TNF-α and IL-6,delay the development of CAVD lesions.
