摘要
为了评价金英黄归汤的抗炎活性,采用Bliss法测定小鼠的半数致死量(LD50)对其进行急性毒性评价,通过体内外试验分析评价其抗炎活性。建立二甲苯致小鼠耳廓肿胀炎症模型,测定小鼠耳廓肿胀度;建立冰醋酸致小鼠腹腔毛细血管通透性炎症模型,测定毛细血管通透性;利用金黄色葡萄球菌建立家兔试验性乳房炎炎症模型,ELISA方法检测乳腺组织和血清中TNF-α、IL-6的分泌量进行抗炎活性评价;利用LPS诱导小鼠乳腺上皮细胞(MECs)建立炎症模型,MTT法检测不同浓度金英黄归汤对MECs细胞活力影响;ELISA方法检测金英黄归汤对LPS诱导MECs的TNF-α、IL-6和IL-1β分泌的影响。结果显示,金英黄归汤灌胃给药对小鼠无明显急性毒性;能显著(P〈0.05)抑制小鼠耳廓肿胀,抑制率为25.42%,能显著(P〈0.05)抑制小鼠腹腔毛细血管通透性,抑制率为33.10%。金英黄归汤各剂量组均能极显著(P〈0.01)降低家兔实验性乳房炎乳腺组织和血清中的TNF-α、IL-6的分泌;金英黄归汤低、中、高(40、400、4000μg/m L)能不同程度的抑制LPS诱导MECs的TNF-α、IL-6和IL-1β的分泌。体内外试验结果表明,金英黄归汤能够对小鼠MECs具有较强的抗炎活性,其抗炎作用与抑制炎性因子的分泌有关。
To evaluate the anti-inflammatory activity of Jinying Huanggui decoction,the half lethal dose(LD50) of mice was examined by Bliss to determine the acute toxicity of the drug. In vivo and in vitro tests were analyzed to evaluate the anti-inflammatory activity of Jinying Huanggui decoction. The in vivo tests included establishing a mouse model of ear edema induced by xylene,acetic acid-induced capillary permeability in mice,and establishing an experimental rabbit breast inflammation model by Staphylococcus aureus. The in vitro test included establishing an inflammation model induced by LPS,detecting the effect of different concentrations of Jinying Huanggui decoction on MEC cells using the MTT method,and measuring the effect of TNF-α,IL-6 and IL-1 β secretion induced LPS using ELISA. The present results showed that there was no obvious acute toxicity to mice by intragastric administration of Jinying Huanggui decoction,the decoction significantly inhibited the ear swelling in mice by 25. 42%(P 0. 05),and significantly inhibited capillary permeability in mice by 33. 10%(P 〈0. 05). Jinying Huanggui decoction significantly reduced the secretion(P 〈0. 01) in rabbits with experimental mastitis breast tissue and serum of TNF-α and IL-6. Jinying Huanggui decoction at low,middle and high concentrations(40,400,and 4000 μg/ml) was nontoxic to MEC cells,and it inhibited the secretion of TNF-α,IL-6 and IL-1β at different degrees. To sum up,we found that Jinying Huanggui decoction possessed a strong anti-inflammatory activity for MECs in mice in in vivo and in vitro tests,which might be related to inhibiting the secretion of inflammatory mediators and cytokines.
