Simultaneous Determination of Nine Marker Components in Yangxinshi Tablet by High performance liquid chromatography

摘要 目的:采用高效液相色谱法同时测定养心氏片中葛根素、大豆苷、丹参素、芦丁、延胡索乙素、淫羊藿苷、芹菜素、人参皂苷Rd、阿魏酸9个成分的含量。方法:采用Agilent Plus-C18色谱柱(250 mm×4.6 mm,5μm),检测波长:阿魏酸采用310 nm,其余8种成分采用254 nm,流动相为乙腈(A)和0.1%甲酸水(B),A相随时间的变化:5%~40%(0~35 min),40%(35~45 min);流速1 m L/min;柱温25℃;进样量5μL。结果:9个指标性成分葛根素、大豆苷、丹参素、芦丁、延胡索乙素、淫羊藿苷、芹菜素、人参皂苷Rd、阿魏酸,在35 min内实现完全分离,线性良好(r>0.9990)。方法学验证表明,日内日间精密度RSD<5%,加样回收率范围为98.91%~102.74%。结论:该方法简便、准确、实用性强,可用于养心氏片的质量控制。 Objective To determine the contents of nine marker components, including puerarin, daidzin, tanshinol, rutin, tetraby- dropalmatine, icariin, apigenin, ginsenoside -Rd, ferulic acid in Yangxinsihi Tablet by high performance liquid chromatography (HPLC) . Methods The HPLC condition was as follows : column: Agilent Plus - C18 (250 mm × 4. 6 mm, 5 μm) ; The detective wave- length was set at 310nm for ferulic acid, and 254 nm for the other eight components. Mobile phase: A was ACN, B was 0. 1% formic acid aqueous solution, the gradient of A phase: 5% -40% (0 -35min), 40% (35 -45min) ; flow rate: 1mL/min; temperature of column: 25℃; injection volume: 51xL. Results Nine marker components, including puerarin, daidzin, tanshinol, rutin, tetrabydropalma- tine, icariin, apigenin, ginsenoside - Rd, ferulic acid, were separated at baseline within 35 min with good linear ( r 〉 0. 9990) . The result of intra-day and inter- day precisions were at normal range (RSD 〈 5% ) . Range of the recovery was between 98.91% and 102. 74%. Conclusion The method is rapid, simple, accurate which can be used to control the quality of Yangxingshi Tablet.
