黄柏提取液对D-氨基半乳糖致小鼠急性肝损伤的预防性保护作用 被引量:9

摘要 目的研究黄柏提取液对D-氨基半乳糖(D-Gal N)致小鼠急性肝损伤的预防性保护作用。方法将60只健康小鼠随机分为正常对照组、模型组、联苯双酯组、黄柏提取液低、中、高三个剂量组,每组10只。联苯双酯组、黄柏提取液各剂量组给予相应的药物灌胃,正常对照组、模型组给予等量生理盐水灌胃,各组小鼠连续灌胃给药10 d,末次给药2 h后,除了正常对照组腹腔注射等量生理盐水外,其他组均按400 mg/kg腹腔注射4%D-Gal N复制急性肝损伤模型。24 h后摘眼球取血,然后处死小鼠取肝组织,比较各组小鼠肝指数、血清AST、ALT水平及血清和肝组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。采用苏木精-伊红染色法观察肝组织的病理变化。结果与正常对照组比较,模型组小鼠肝脏指数及血清ALT、AST水平升高,血清和肝组织中SOD活性降低,MDA含量增高,肝组织损伤严重。与模型组比较,黄柏提取液的低、中、高剂量组及联苯双酯组小鼠肝脏指数及血清ALT、AST水平降低,黄柏提取液的中、高剂量组及联苯双酯组血清和肝组织中SOD活性升高,MDA含量降低,肝组织损伤有明显好转。结论黄柏提取液对D-Gal N诱导的急性肝损伤有较好的预防性保护作用。 Objective To study the preventive and protective effects of Cortex Phellodendri extracts on mice with acute liver injury induced by D-galactosamine( D-Gal N). Methods Sixty healthy mice were randomly divided into normal control group,model group,bifendate group,low-,medium-and high-dose Cortex Phellodendri extracts groups,with 10 mice in each group. The bifendate group and the Cortex Phellodendri extracts groups were given corresponding drugs by gavage,while the normal control group and the model group were given an equivalent volume of normal saline by gavage. The mice in each group received the consecutive administration by gavage for 10 days. After two hours of the final administration,except the normal control group was intraperitoneally injected with an equivalent volume of normal saline,all groups with 4% D-Gal N( 400 mg/kg) to replicate the model of acute liver injury. Twenty-four hours later,the eyeballs of mice were removed and blood samples were collected,then the mice were sacrificed for liver tissues. The liver index,the levels of serum AST and ALT,the activity of superoxide dismutase( SOD) and the content of malondialdehyde( MDA) in serum and liver tissues were compared between the two groups. Hematoxylin-eosin staining was used to observe the pathological changes in liver tissues. Results Compared to the normal control group,the mice in the model group obtained increased liver index,elevated levels of serum ALT and AST,decreased activity of SOD in serum and liver tissues,and increased MDA content,and presented a severe liver tissue damage. Compared to the model group,the liver index and the levels of serum ALT and AST decreased in the low-,medium-and high-dose Cortex Phellodendri extracts groups and the bifendate group,the serum and liver tissues SOD activity increased but the MDA content decreased in the medium-and high-dose Cortex Phellodendri extracts groups and the bifendate group,and liver tissue damages improved markedly. Conclusion Cortex Phellodendri extracts has better preventive and protective effects on acute liver injury induced by D-Gal N.
