摘要
目的建立HPLC波长切换法同时测定茵陈五苓糖浆中滨蒿内酯、金丝桃苷、异槲皮苷、苍术素、24-乙酰泽泻醇A和23-乙酰泽泻醇B含量。方法采用Diamonsil C_(18)色谱柱(250 mm×4.6 mm,5μm);以乙腈-0.1%磷酸溶液为流动相,进行梯度洗脱;流速0.9 mL·min^(-1);检测波长:345 nm(滨蒿内酯、金丝桃苷、异槲皮苷和苍术素)、208 nm(24-乙酰泽泻醇A和23-乙酰泽泻醇B);柱温30℃。结果滨蒿内酯、金丝桃苷、异槲皮苷、苍术素、24-乙酰泽泻醇A和23-乙酰泽泻醇B的线性范围分别为17.63~440.75μg·mL^(-1)(r=0.9993)、4.89~122.25μg·mL^(-1)(r=0.9998)、2.47~61.75μg·mL^(-1)(r=0.9996)、9.66~241.50μg·mL^(-1)(r=0.9991)、1.95~48.75μg·mL^(-1)(r=0.9999)、3.37~84.25μg·mL^(-1)(r=0.9995),平均加样回收率均大于95%,RSDs<2.2%。结论本实验建立的茵陈五苓糖浆的多成分测定方法可靠,重复性好,可用于茵陈五苓糖浆的质量控制。
Objective To simultaneously determine scoparone, hyperoside, isoquercitrin, atractylodin, 24-acetate alisol A and 23-acetate alisol B in Yinchen Wuling syrup by HPLC wavelength switching method. Methods The Diamonsil C18 (250 mm× 4.6 mm, 5μm) chromatographic column was adopted. The mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution with gradient elution at a flow rate of 0.9 mL · min^-1. The detection wavelength was 345 nm (scoparone, hyperoside, isoquercitrin and atractylodin) and 208 nm (24-acetate alisol A and 23-acetate alisol B). The column temperature was 30℃. Results The calibration curves of scoparone, hyperoside, isoquercitrin, atractylodin, 24-acetate alisol A and 23-acetate alisol B were linear at 17.63 - 440.75μg·mL^-1 (r = 0.9993), 4.89 -- 122.25μg·mL^-1 (r = 0.9998), 2.47 - 61.75μg·mL^-1 (r = 0.9996), 9.66 - 241.50 μg·mL^-1(r =0.9991), 1.95 - 48.75μg·mL^-1 (r = 0.9999), and 3.37 - 84.25μg·mL^-1(r =0.9995). The average recoveries were higher than 95%, RSDs 〈 2.2%. Conclusion The method is reliable and reproducible, which can be used for the quality control ofYinchen Wuling syrup.
