摘要
目的探讨慢性心衰大鼠心肌细胞凋亡、心肌纤维化及其相互关系。方法清洁级SD大鼠20只,体重250~300 g,随机分为心衰组及正常组,每组10只。心衰组腹腔连续注射异丙肾上腺素建立大鼠慢性心衰模型。造模6 w后,麻醉大鼠,剪开胸腔,取左心室心肌组织标本,伊红-苏木素(HE)染色,观察心肌组织病理变化,Masson染色显示心肌组织中胶原纤维化情况,TUNEL染色检测心肌细胞凋亡率。结果心衰组心肌纤维化程度较正常组显著增加,且发生心肌细胞肥大及坏死、心肌纤维断裂、心肌间质纤维化瘢痕组织形成、心肌组织结构紊乱;心衰组心肌细胞凋亡率较正常组增高(P<0.01),且与心肌胶原容积比呈显著正相关(r=0.895,P<0.01)。结论心肌细胞凋亡及心肌纤维化是慢性心衰大鼠的重要病理改变,且两者呈显著正相关。
Objective To explore the cardiomyocyte apoptosis and the myocardial fibrosis in rats with chronic heart failure and their correlation. Methods 20 clean sprague-dawley (SD) rats weighing 250-300 g were selected and randomly divided in to a heart failure model group and a normal control group, with 10 rats in each group. The heart failure group received continuous intraperitoneal injection of isoproterenol to build a rat model of chronic heart failure. Six weeks after the modeling, the rats were anesthetized and their chest were cut open; specimens were obtained from their left ventricular myocardia; the hematoxylin-eosin (H-E) staining was performed to observe the pathological changes of the myocardia; the Masson's trichrome staining was performed to show the fibrosis of collagen in the myocardia; and the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) method was conducted to measure the cardiomyocyte apoptosis. Results The myocardial fibrosis obviously became more severe in the heart failure group as compared with that in the normal group; the hypertrophy and necrosis of cardiomyocytes, the breakage of myocardial fibers, the myocardial interstitial fibrosis and formation of cicatricial tissues, and the structural disorder in myocardia were discovered in the heart failure group; the cardiomyocyte apoptosis rate in the heart failure group increased as compared with that of the normal group (P 〈 0.01); and the cardiomyocyte apoptosis rate was significantly and positively correlated with the collagen volume fraction (r= 0.895, P 〈 0.01). Conclusion The cardiomyocyte apoptosis and myocardial fibrosis are important pathological changes in rats with chronic heart failure; they are significantly and positively correlated with each other.
