摘要
目的观察沉默CD98hc对乳腺癌MDA-MB-231细胞体外增殖、迁移及侵袭的影响。方法应用RNA干扰技术沉默乳腺癌MDA-MB-231细胞CD98hc的表达,筛选并获得稳定CD98hc低表达的MDA-MB-231细胞株(CD98hc-shRNA)及对照组细胞(Vector)。应用Western blot及qRT-PCR技术鉴定CD98hc的表达;应用MTT细胞增殖实验检测细胞增殖能力的改变;细胞迁移侵袭实验检测细胞迁移和侵袭能力的改变。结果 CD98hc-shRNA下调乳腺癌MDA-MB-231细胞CD98hc的表达,与空白组(0.706±0.013)、对照组(0.724±0.018)相比,实验组(0.580±0.035)细胞的增殖能力明显下调(P<0.05),且迁移和侵袭能力也显著受抑制(P<0.05),空白组与对照组差异无统计学意义(P>0.05)。结论 CD98hc可能参与乳腺癌细胞生物学行为的调控。
Purpose To investigate the effect of silencing of CD98bc on proliferation, migration and invasion of MDA-MB- 231 breast cancer cells. Methods RNA interfere technology was used to silence CD98hc in MDA-MB-231 cells. MDA-MB- 231 cells (CD98he-shRNA) with stable low expression of CD98hc and Vector control was obtained. Western blot and qRT-PCR were used to verify the down-regulation of CD98hc. MTT assay was used to test the cell proliferation. Transwell mi- gration and invasion experiment were used to assay cell migration and invasion. Results The expression levels of CD98hc was down-regulated by CD98hc-shRNA in the MDA-MB-231 breast cancer cells. Compared to that of blank cells (0. 706 ±0. 013) , vector control (0. 724 ±0.018 ), the cell proliferation potency of CD98hc-shRNA cells (0. 580 ±0. 035) was significantly inhibi- ted ( P 〈 0. 05 ) , and the cell migration and invasion potency al- so were inhibited ( P 〈 0. 05 ), there is on significant different between blank cells and vector control ( P 〈 0. 05 ). Conclusion CD98hc might be involved in the regulation of breast cancer cell biological behaviors.
