Determination of Seven Kinds of Phenylethanol Glycosides in Gesneriaceae Species from Guilin Habitats by UPLC-MS/MS

摘要 目的:建立UPLC-MS/MS同时测定苦苣苔科植物中7个苯乙醇苷类成分的方法,分析比较桂林产半蒴苣苔属植物中苯乙醇苷含量差异。方法:采用ACQUITY UPLC BEH C18色谱柱(100 mm×2.1 mm,1.8μm),以0.1%甲酸溶液(A)-0.1%甲酸乙腈溶液(B)为流动相,梯度洗脱,流速为0.6 m L/min;柱温40℃;采用电喷雾离子源,负离子检测方式,得到相应的提取离子流图,以峰面积进行定量。结果:7个苯乙醇苷类成分具有良好的线性关系,r值均大于0.9990,方法精密度、重复性和稳定性的RSD值均小于3.5%,加样回收率在97.1%~103.2%,RSD均小于3.6%。14种半蒴苣苔属植物中7个苯乙醇苷在组成和含量上有一定差异。结论:该方法操作简便、准确、重复性好,对寻找高含量苯乙醇苷类资源植物具有指导意义。 Objective:To establish an UPLC-MS/MS method for the simultaneous determination of seven phenylethanol glycosides in Gesneriaceae species, and to analyze the difference of phenylethanol glycosides ingredients in the samples from Guilin habitats. Methods:The determination was performed on the AQUITY BEH Cls (100 mm × 2. 1 mm, 1.8 μm) ;the mobile phase was 0. 1% formic acid (A) -0. 1% formic acid acetonitrile (B) in gradient elution; the flow rate was 0. 6 mL/min; the column temperature was 40 ℃. MS instrument was equipped with ESI-ion source. Gaining the extracted ion chromatograms, then the peak area was used for quantitative. Results: Seven phenylethanol glycosides showed good linearity( r ≥0. 9990). The RSD of the precision, repeatability, and stability tests were less than 3.5%. The average recovery rates were in the range of 97. 1% -103. 2% ,the RSD were less than 3.6%. There are differences in the composition and content of seven phenylethanol glycosides from different Gesneriaceae species. Conclusion :The method for the sim- ultaneous determination of the seven phenylethanol glycosides is simple, accurate, and reproducible,which has great theoretical signifi- cance for looking for high levels of phenylethanol glycosides of plant resources.
