Optimization of fermentation conditions for Dioscorea zingiberensis dioscin hydrolase production

摘要 从盾叶薯蓣根茎清洗液中,筛选出一株产薯蓣皂苷水解酶菌株Aspergillus sp.,对该菌株产酶发酵条件进行优化研究。单因素试验结果表明,0.3%葡萄糖为碳源、0.4%蛋白胨为氮源,薯蓣皂苷水解酶活力为0.78 U/m L。正交试验结果表明,蛋白胨含量对酶活力的影响较大,产酶发酵培养基最佳配方为0.5%葡萄糖,0.6%蛋白胨,0.2%薯蓣皂苷,0.1%K_2HPO_4,0.05%MgSO_4·7H_2O,0.05%KCl,0.001%FeSO_4·7H_2O。在此最佳培养基配方条件下,薯蓣皂苷水解酶活力为0.96 U/m L。0.2%麦冬、0.2%绞股蓝、0.2%薯蓣三种混合皂苷作为诱导物时,薯蓣皂苷水解酶活力最高,达1.26 U/m L。 Aspergillus sp. with dioscin hydrolase activity was isolated from the rhizosphere of Dioscorea zingiberensis. The optimization of fermentation conditions for the dioscin hydrolase production by Aspergillus sp. was studied in this paper. Single factor experiments results showed that dioscin hydrolase activity was 0.78 U/ml with glucose 0.3% as carbon source and peptone 0.4% as nitrogen source. The orthogonal experiments results showed that peptone concentration was the primary factor affecting dioscin hydrolase activity. The optimum formula of fermentation medium was glucose 0.5%, peptone 0.6%, dioscin 0.2%, K2HPO4 0.1%, MgSO4·TH2O 0.05%, KCl 0.05%, FeSO4·7H2O 0.001%. The strain producing dioscin hydrolase was greatly induced by the mixed dioscin, and the activity reaching 1.26 U/ml.
