摘要
目的:探讨泰山磐石散对反复自然流产小鼠母胎界面IFN-γ、IL-10的含量、SOCS1及SOCS3的基因表达的影响。方法:采用雌性CBA/J与雄性BALB/c小鼠交配建立正常妊娠小鼠模型,采用雌性CBA/J小鼠与雄性DBA/2小鼠交配建立反复自然流产小鼠模型,并将流产模型小鼠随机分为4组,分别为反复自然流产模型组及泰山磐石散低、中、高剂量组。泰山磐石散低、中、高剂量组从合笼开始灌胃给药,灌胃治疗14 d后处死小鼠,取其蜕膜和胎盘组织,Elisa技术测定分别检测IFN-γ、IL-10的含量,采用荧光定量PCR法测定各组小鼠蜕膜及胎盘组织中SOCS1及SOCS3的基因表达。结果:小鼠组织中IL-10的含量,模型组较正常组显著降低(P<0.05),IFN-γ的含量,模型组较正常组显著升高(P<0.05),经过中药干预后,与模型组相比较,中、高剂量组IL-10的含量明显升高(P<0.05),IFN-γ的含量明显下降(P<0.05);小鼠组织中SOCS1表达量,模型组较正常组显著升高(P<0.05),SOCS3表达量,模型组较正常组显著降低(P<0.05),经过中药干预后,SOCS1表达量均降低,中、高剂量组与与模型组比较有显著性差异(P<0.05),各剂量组SOCS3的表达量呈不同程度的升高,高剂量组与模型组比较有显著性差异(P<0.05)。结论:泰山磐石散可能通过降低IFN-γ的含量、提高IL-10的含量,同时通过降低小鼠母胎界面SOCS1蛋白表达、提高SOCS3蛋白表达,进而调控Th细胞向Th2方向分化,故对反复自然流产有治疗作用。
Objective : To study the effect of Taishan Panshi Powder on contents of IFN - γ and IL - 1 0 as well as SOCS1 and SOCS3 gene expressions of maternal - fetal interface of repeated spontaneous abortion mice. Methods : CBA/ J females and males mate BALB/c mice were used to establish the normal pregnancy mice model. Use the female CBA/ J mice anf male DBA/2 mice mating to set up the repeated spontaneous abortion mice model. The abortion model mice were randomly divided into 4 groups: repeated spontaneous abortion model group and Taishan Panshi Powder low, medium and high doses groups. Taishan Panshi Powder low, medium and high doses groups were given the medicine starting from the close cage. After 14 days, execute the mice and take out decidua and placental tissue. ELISA was used to detect the contents of IFN -γ, and IL- 10. By fluorescence quantitative PCR method, decidua and placental tissue SOCS1 and SOCS3 gene expressions were detected. Results : The content of IL - 10 in the model group decreased significantly compared with that of the normal group (P 〈 0. 05 ) and the content of IFN - γ, iin the model group significantly inc,eased compared with that of the normal group ( P 〈 0. 05 ). After the intervention, compared with model group, middle and high dose groups' IL - 10 levels increased significantly ( P 〈 0. 05 ). The content of IFN -γ significantly decreased ( P 〈 0. 05 ). SOCS1 expression in the model group significantly increased compared with that of the normal group ( P 〈 0. 05). The amount of SOCS3 expression in the model group significantly reduced compared with that in the normal group ( P 〈 0. 05 ). After the intervention, the expression of SOCS1 quantity was lower. The middle and high dose groups had significant difference compared with the model group (P 〈 0. 05 ). The expression of each dose group' s SOCS3 quantity in different degree increased. The high dose group had significant difference compared with model group(P 〈0. 05). Conclusion: Taishan Panshi Powder may controls Th cells Th2 direction differentiation by lowering the content of IFN - γ and improving the content of IL - 10, at the same time by reducing the maternal - fetal interface SOCS1 protein expression in mice and improving SOCS3 protein expression, and thus has therapeutic effect on repeated spontaneous abortion.
