摘要
为了评价混合青贮的品质和筛选优良的乳酸菌菌株,本研究以不同比例(甜高粱与苜蓿比例分别为0∶100,20∶80,40∶60,60∶40,80∶20,100∶0)的甜高粱与苜蓿混合青贮为材料,对混合青贮进行了感官评价和发酵品质分析,通过MRS平板分离、生理生化分析和16S rRNA相结合的方法对不同比例混合青贮中乳酸菌的种类、生理生化特性进行了研究。结果表明,40%SS比例的混合青贮效果最佳。从不同比例混合青贮中共分离得到18株乳酸菌,4株鼠李糖乳杆菌(Lactobacillus rhamnosus),5株植物乳杆菌(Lactobacillus plantarum),3株耐久肠球菌(Enterococcus durans),2株肠膜明串珠菌肠膜亚种(Leuconstoc mesenteroides subsp mesenteroides),2株屎肠球菌(Enterococcus faecium),乳酸乳球菌(Lactococcus lactis)和干酪乳杆菌(Lactobacillus casei)各1株。各菌株产酸试验和生长试验结果表明,植物乳杆菌产酸性能好,生长速度快,可作为制作混合青贮饲料的优良菌株。
In order to evaluate and screen fine strains of lactic acid bacteria silage quality, study used silages with different proportions of sweet sorghum mixed with alfalfa( Sweet sorghum and alfalfa proportion respectively0:100 20:80 40:60 60:40 80:20 100: 0) as ma- terials to investigate the silages sensory profile, fermentation quality as well as the variety and physiological - biochemical characteristic of lactic acid bacteria in the silages using the methods of bacteria culture and 16S rRNA technology. The results showed that the highest quality was the mixed silage including 40% sweet sorghum, and a total number of 18 lactic acid bacteria, which composed of 4 strains of Lactobacillus rhamnosus, 5 strains of Lactobacillus plantarum, 3 strains of Enterococcus durans, 2 strains of Leueonstoc mesente- roides subsp mesenteroides, 2 strains of Enterocoecus faecium, and 1 strain of Lactoeoccus lactis and Lactobacillus casei seperatively, were isolated from the mixed silages, results of acid production and growth test of the different strains showed that plant lactobacillus produced the most amount of acid and grew the most quickly compared with any other bacterial strains, suggesting that Lactobacillus Dlantarum could be a fine strain and has potential value for silage making.
