摘要
目的探讨健脾摄血方对ITP模型小鼠血清β-内啡肽(β-EP)、血管活性肠肽(VIP)、5-羟色胺(5-HT)、去甲肾上腺素(NE)、缩胆囊素(CCK)的影响。方法以免疫造模法建立ITP小鼠模型,以强的松、健脾摄血方为干预药物,用酶联免疫吸附法(ELISA)检测模型小鼠血清β-EP、VIP、5-HT、NE、CCK含量变化。结果 (1)给药第8天,与空白对照组比较,模型对照组小鼠外周血小板计数仍低于正常水平(P<0.01);强的松对照组、健脾摄血组血小板计数明显提升,与模型对照组比较,差异有统计学意义(P<0.01)。(2)注射APS前、后各时间段,与空白对照组比较,实验各组小鼠PDW无明显变化(P>0.05)。(3)β-EP:与空白对照组比较,模型对照组小鼠血清β-EP含量升高,差异有统计学意义(P<0.05);与模型对照组比较,强的松干预后小鼠血清β-EP含量变化差异无统计学意义(P>0.05),健脾摄血方干预后,差异有统计学意义(P<0.05)。(4)VIP:与空白对照组比较,模型对照组VIP含量明显降低,差异有统计学意义(P<0.05);与模型对照组比较,强的松对照组、健脾摄血组VIP含量明显回升,差异有统计学意义(P<0.05)。(5)5-HT:与空白对照组比较,模型对照组5-HT含量显著降低,差异有统计学意义(P<0.05);与模型对照组比较,强的松对照组(P<0.01)、健脾摄血组(P<0.05)含量显著回升。(6)NE:与空白对照组比较,模型对照组、强的松对照组、健脾摄血组小鼠血清NE含量变化差异有统计学意义(P<0.05)。(7)CCK:与空白对照组比较,模型对照组、强的松对照组、健脾摄血组小鼠CCK含量变化差异均无统计学意义(P>0.05)。结论免疫造模法能够成功复制ITP小鼠模型,健脾摄血方除能够提升ITP模型小鼠外周血小板数值外,对ITP模型小鼠血清β-EP、VIP、5-HT、NE含量有明显的调控效应。
Objective To explore the impacts of jianpi shexue formula on serum β-EP,VIP,5-HT,NE and CCK in ITP model mice.Methods The immune modeling method was used to establish ITP model mice.Prednisone and jianpi shexue formula were used in the intervention.ELISA was adopted to determine the levels of β-EP,VIP,5-HT,NE and CCK.Results 1.On the 8th day of medication,compared with the blank control group,the peripheral platelet count in the mice of the model control group was still lower than the normal level(P〈0.01).the platelet counts in the prednisone control group and the jianpi shexue group were increased apparently,indicating the significant difference as compared with the model control group(P〈0.01).2.Before APS injection and at each time stage after injection,compared with the blank control group,PDW did not change apparently in each experimental group(P〉0.05).3.β-EP:compared with the blank control group,β-EP level was increased in the model control group,indicating the significant difference(P〈0.05).Compared with the model control group,after the intervention with prednisone,β-EP level did not change significantly(P〉0.05).After the intervention with jianpi shexue formula,the difference was significant statistically(P〈0.05).4.VIP:compared with the blank control group,VIP level in the model control group was reduced apparently,indicating the significant difference(P〈0.05).Compared with the model control group,VIP level was increased apparently in the prednisone group and jianpi shexue group,indicating the significant difference(P〈0.05).5.5-HT,compared with the blank control group,5-HT was reduced significantly in the model control group,indicating the significant difference(P〈0.05).Compared with the model control group,it was increased significantly in the prednisone group(P〈0.01)and jianpi shexue group(P〈0.05).6.NE:compared with the blank control group,the difference in NE level was significant in the model control group,the prednisone group and jianpi shexue group(P〈0.05).7.CCK:compared with the blank control group,the difference in CCK level was not significant in the model control group,the prednisone group and jianpi shexue group(P〉0.05).Conclusion The immune modeling method duplicates successfully ITP model mice.Jianpi shexue formula increases peripheral platelet counts in ITP model mice and apparently regulates the levels of β-EP,VIP,5-HT,NE and CCK in serum of IPT model mice.
