水杨酸和普鲁兰多糖对东北刺人参不定根生物活性物质积累的影响 被引量:1

摘要 诱导子是促进植物细胞、组织和器官培养物中生物活性物质最常用的方法。本试验将水杨酸(SA)和普鲁兰多糖(PL)作为2种诱导子分别对东北刺人参不定根进行处理,旨在探究2种诱导子对悬浮培养的东北刺人参不定根中多糖、酚类及黄酮等生物活性物质积累的影响。同时,对东北刺人参不定根的乙醇提取物的抗氧化特性进行了研究。结果表明,经SA、PL处理后,不定根中生物活性物质的积累量显著增加。在悬浮培养40d时加入100!mol SA处理4d时多糖、酚类和黄酮的积累量达到最大值,产量分别比未处理组高出1.22、1.45和1.45倍;PL在200mg/L时诱导效果最佳,且相比酚类和黄酮,多糖积累量增加最为显著。对于东北刺人参不定根提取物的抗氧化能力的测定结果显示,DPPH及烷基自由基的ESR信号强度随着提取物浓度的增加而降低。 Alicitors are commonly used to promote bioactive compounds in plant cells,tissues and organ cultures.In the present study,salicylic acid(SA)and pullulan(PL)were used as two elicitors to deal with the adventitious roots of the Oplopanax elatus,respectively.The aim of this study was to investigate the effects of two elicitors on the accumulation of polysaccharides,phenol and flavonoids.At the same time,the antioxidant properties of adventitious ethanol extract were studied.The results showed that the accumulation of bioactive compounds in adventitious roots increased in SA treatment significantly.The accumulation of polysaccharides,phenol and flavonoids reached the maximum levels at the time of 40 days of suspension treatment,and the yields were 1.22,1.45,1.45 times higher than that of the untreated group,respectively.PL was the best at 200mg/L,and the accumulation of polysaccharides was the most significant.The results of the determination of the antioxidant capacity of the adventitious root extract showedthat the ESR signal strength of the 1,1-diphenyl-2-picrylhy-drazyl(DPPH)and alkyl radicals obviously decreased with the increasing adventitious roots extract concentrations.
