摘要
采用Penomenex氨基柱为分析柱,建立了高效液相色谱法-蒸发光散射检测方法(HPLC-ELSD)测定油梨种子中不同可溶性糖的方法,并比较了热处理对油梨种子中不同可溶性糖的含量的影响。高效液相色谱分析条件的优化条件为乙腈-水(88:12,体积比)为流动相,流速1.0 mL/min,柱温25℃,氮气压力为172 kPa,漂移管温度为60℃。6种糖标准品线性关系良好。检出限为0.09~0.41 mg/L,样品的加标回收率84.5%~104.7%,相对标准偏差(RSD)小于4.9%。油梨种子中的可溶性糖主要由果糖、葡萄糖、D-甘露庚酮糖和甘露庚糖醇组成;油梨种子经热处理后,其中可溶性糖含量均有所下降。
A new approach for the determination of different soluble saccharides from avocados seed is described, and effects of heat treatment on the contents of soluble saccharides are investigated. The extracts were analysed by high performance liquid chromatography with evaporative light scattering detector (ELSD). The soluble saccharides were separated on Penomenex amino column (25 ℃ ) using an acetonitrile-water gradient system at a flow-rate of 1.0 mL/min. The optimum setting of the ELSD parameters: drift tube temperature was 60 ℃ and nitrogen cartier gas pressure was 172 kPa The linearity of six soluble saccharides standards was good. The detection limits were in the range of 0. 09-0. 41 mg/L, and the recoveries ranged from 84. 5% to 104. 7% with deviations (RSD) 〈 4.9%. The soluble saccharides in avocado seed were the relative standard mainly composed of fructose, glucose, D-mannoheptulose and perseitol, and the contents decreased after heat treatment. The method is accurate, reproducible and has been applied for quantitative analysis of soluble saccharides in avocados seed.
