摘要
目的观察苦豆碱对膀胱癌EJ细胞增殖与凋亡的影响并探讨其作用机制。方法实验分组如下,A组:膀胱癌EJ细胞不作处理;B组:加入苦豆碱浓度25μmol/L;C组:加入苦豆碱浓度50μmol/L;D组:加入苦豆碱浓度100μmol/L。通过CCK8检测各组细胞的存活率,通过流式细胞术检测各组细胞凋亡率,通过Western blot法检测各组细胞Bcl-2、Bax、p-Erk1/2蛋白的表达水平。结果 CCK8结果显示,B、C、D组较A组细胞存活率降低(P<0.05);流式细胞术结果显示:B、C、D组较A组细胞凋亡率增加(P<0.05);Western blot结果显示:B、C、D组较A组Bcl-2、p-Erk1/2表达水平降低(P<0.05),Bax表达水平增高(P<0.05)。结论苦豆碱可降低膀胱癌EJ的存活率,诱导其凋亡,其作用机制可能是通过抑制Erk1/2的磷酸化而实现的。
Objective To investigate the effect of aloperine on the proliferation and apoptosis of bladder cancer EJ cells and to ex- plore its mechanism. Methods The experimental group was as follows: bladder cancer EJ cells were not treated (group A) ; 25μmlol/L aloperine was added (group B) ; 50μmol/L aloperine was added (group C). 100μmol/L aloperine was added (group D). The survival rate of the cells was detected by CCK8. The apoptosis rate was detected by flow cytometry. The expression levels of Bel - 2, Bax, p - Erkl/2 protein were detected by Western blot. Results CCK8 results showed that: B, C, D group compared with A group, the cell sur- vival rate were decreased (P 〈 0.05). The results of flow cytometry showed that the apoptosis rate of B, C and D group was higher than that of A group (P 〈 0.05 ). Western blot results showed that : B, C, D group compared with A group the expression of Bcl - 2 and p - Erkl/2 levels were reduced (P 〈 0.05) , the expression of Bax level was increased (P 〈 0.05 ). Conclusion The survival rate of EJ in bladder cancer can be reduced by aloperine and induce its apoptosis, and inhibiting the phosphorylation of Erkl/2 may be the mechanism.
