基于环糊精构建的超分子基因输送载体

Cyclodextrin-Based Supramolecular Vehicles for Gene Delivery

摘要 基因治疗是对付恶性肿瘤的有效手段。然而,缺乏高效和安全的基因输送载体限制了它在临床上的使用。制备药物输送载体是生物大分子非常重要的用途。超分子是生物大分子家族的重要成员,近来的研究发现基于环糊精构造的超分子能有效传送基因至癌细胞内,并实现高效转染。通过参阅近3年来的文献,作者概述了这类超分子作为基因载体的研究进展,显示这类新兴载体的应用价值和前景。 Supramolecular systems are grounded on weak and reversiblenon-covalent interactions, such as hydrogen bonding, metal coordination,hydrophobic attractions, and electrostatic interactions. Recent advancements in supramolecular chemistry have lately aroused a growing interest to self-assembled supramolecular blocks in the pharmaceutical applications. Therapeutic genes can be tem- porarilyen capsulated into functional supramolecules and released at a specific target or with designated timing. Cyclodextrin-based carriers for gcne delivery have not only optimal drug loading capacities, release properties and the ease for the incorporation of muhi- ple functionalities, but also contain biocompatible macromolecular backbones and receive smart responsiveness to various physio- chemical stimuli,thereby holding great promising for a wide application in gene therapy. Many polycationic types of cyclodextrins have already been developed for supramolecular formulations. In this feature article ,we overview the recently reported supramolecular systems derivin- from cvclodextrins for -ene delivery.
