摘要
目的:研究参芪扶正注射液联合顺铂对人胰腺癌PANC-1细胞增殖、诱导凋亡的影响,并讨论其作用机制。方法:常规培养细胞,采用参芪扶正注射液和顺铂分别单独及联合作用于PANC-1细胞,于倒置显微镜下观察细胞形态的变化;MTT法检测不同浓度的参芪扶正注射液和顺铂单独及联合作用PANC-1细胞后的抑制率;流式细胞术检测细胞凋亡率,ELISA法检测凋亡因子Caspase-9、Fas和Survivin的含量。结果:参芪扶正注射液和顺铂均能有效抑制PANC-1细胞增殖,并诱导其凋亡,两药联合应用具有协同效应,可促进Caspase-9、Fas的表达,抑制Survivin的表达。结论:参芪扶正注射液联合顺铂能协同抑制PANC-1细胞的增殖,并诱导其凋亡,其作用机制与上调Caspase-9、Fas及下调Survivin因子的表达有关。
Objective： To investigate the effect and mechanism of Shenqi Fuzheng injection combined with cisplatin on proliferation and apoptosis of pancreatic cancer cell line PANC-1. Methods： Cells were cultivated in vitro; PANC-1 cells were treated with Shenqi Fuzheng injection alone or combined with cisplatin. Cellular morphologic changes were observed under inverted phase contrast microscope. The effects of proliferation inhibition were assayed by M3T. The combination effect was judged through Chou-Talaly analysis. The apoptosis ratios of ceils were analyzed by flow cytometry.The gene expression of Caspase-9, Fas and Survivin was detected by ELISA. Results： Shenqi Fuzheng injection alone or combined with cisplatin could inhibit the proliferation, and induce the apoptosis of PANC-1 cells. Combination of Shenqi Fuzheng injection and cisplatin revealed a synergistic effect on cell proliferation and apoptosis as it enhanced the expression of Caspase-9 and Fas but reduced that of Survivin. Conclusion： Synergistic effect of Shenqi Fuzheng injection combined with cisplatin on cell proliferation and apoptosis of PANC-1 ceils is possibly due to enhancement of Caspase-9, Fas and reduction of Survivin expression.
