摘要
目的建立鉴别老鼠瓜凝胶剂中槲皮素的薄层色谱法(TLC)和测定盐酸水苏碱含量的高效液相色谱法(HPLC)。方法采用薄层谱法对老鼠瓜凝胶剂中槲皮素进行了鉴别,以甲苯-乙酸乙酯-甲酸(5∶2∶1)为展开剂。采用高效液相色谱法测定老鼠瓜凝胶剂中盐酸水苏碱的含量,色谱柱为C18柱(250mm×4.6 mm,5μm),流动相为乙腈-水=80∶20,流速为0.8mL/min,检测波长202nm,柱温20℃,进样量10μL。结果在薄层色谱中能检测到槲皮素,薄层展开后均显示与槲皮素对照品相同的斑点。盐酸水苏碱质量浓度在0.1~1.0 mg/mL范围内线性良好,样品平均回收率为99.8%(n=6),RSD为0.7%,盐酸水苏碱的含量为0.281~0.440mg/g,平均含量为0.357mg/g。结论本研究建立的定性和定量方法稳定、可靠、专属性强,可用于维药老鼠瓜凝胶剂中的槲皮素的定性鉴别和盐酸水苏碱的含量测定。
Objective To establish the thin layer chromatography (TLC) identification method of quercetine and HPLC content determination method of stachydrine hydrochloride in Uyghur medicine Capparis spinosagels. Methods Quercetinin extracted from Uyghur medicine Capparis spinosagels was i- dentified by TLC. The content of stachydrine hydrichloride in Uyghur medicine Capparis spinosa gels was determined by HPLC method. The HPLC conditions used was C18 column (4.6 mm× 250 mm, 5 μm), mobile phase was acetonitrile-water (80 : 20), detection wavelength was 202 nm, flow rate was 0.8 mL/min, column temperature was 20℃, the injection volume was 10 μl. Results Quercetin could be detected by TLC method, the principal spot in the chromatogram obtained with test solution was similar to the princi- pal spot in the chromatogram obtained with the reference solution. There was good linear correlation in the range of 0.1-1.0 mg/ml in the case of determination of stachydrine hydrochloride, the average recovery was 99.8% with RSD of 0.7% (n =6),the content of stachydrine hydrochloride was 0.281-0.440 mg/g. Conclusion The method we established was accurate, reliable and specific and was shown to be able to be used for identification of quercetin and content determination of stachydrine hydrochloride in Uyghur medi- cine Capparis spinosa gels.
