摘要 目的探索能通过流式细胞术快速测定细胞周期的方法.方法培养的小鼠成纤维细胞分别采用DNA一步法和溴化丙啶(PI)标准染色流程染色,应用流式细胞仪检测细胞G_1期、S期和G_2/M期的DNA含量,比较2种方法的测定结果.结果 2种染色方法标记的小鼠成纤维细胞呈现完整的细胞周期峰,G_1含量分别为65.85%和66.54%,S期含量分别为13.45%和13.32%,G_2/M期含量分别为20.71%和20.15%,结果差异无统计学意义(P>0.05).结论 DNA一步法能通过流式细胞术快速准确地测定细胞周期. Objective To explore the method that can rapidly analyze cell cycle by flow cytometer. Methods Mouse fibroblast cells stained with DNA one-step method and Bromide (PI) standard dyeing process were measured by flow cytometer for the percentages of cells in Gl, S and GJM phases. The results were analyzed and compared. Result The complete cell cycle of mouse fibroblast cells stained with DNA one-step method or Bromide bromide (PI) standard dyeing process was showed entirely, the percentages of cells in G1 were 65.85% and 66.54%, in S were 13.45% and 13.32%, and in GJM, 20.71% and 20.15%, respectively. The results from the two different methods showed no significant difference (P〉0.05) . Conclusions The DNA one-step method can quickly and accurately analyze cell cycle by flow cytometer.
