气调包装对鲳鱼冷藏期间主要微生物多样性的影响 被引量:11

Effect of Modified Atmosphere Packaging (MAP) on the Microbial Diversity Changes of Pomfret (Pampus argenteus) during Chilled Storage

摘要 为研究气调包装对冷藏鲳鱼贮藏期间主要微生物多样性的影响,分别选取不同时间的对照组与处理组样品,以感官评定、pH值与细菌总数为评价指标,进行微生物的分离纯化,提取细菌DNA,并作PCR扩增。测序后进行相似性比对,辅以生理生化鉴定其种类分布与数量变化。结果表明:采用80%CO_^(2+)10%O_^(2+)10%N2的气调包装处理,能延缓鲳鱼鲜度的下降,抑制微生物的生长,使鲳鱼的冷藏货架期延长3~4 d。经生理生化鉴定、形态学观察与PCR扩增测序,最终得到6株具有典型特征的纯菌株。贮藏前期,革兰氏阴性菌为主要优势菌,而贮藏后期,气调处理对革兰氏阴性菌的生长有明显抑制。腐败希瓦氏菌、荧光假单胞菌与嗜冷杆菌为鲳鱼冷藏期间的主要优势菌。同时,还有一定比例的热杀索丝菌与草莓假单胞菌。 To investigate the effect of modified atmosphere packaging (MAP) to the microbial diversity changes of pomfret (Pampus argenteus) during chilled storage.Sensory evaluation,pH value and total viable counts (TVC) was measured at the controlled group and MAP group at different storage time.Followed by sequencing which were aligned to 16S rDNA gene fragments available from NCBI to find the closest known relatives to this partial 16S rDNA sequences.The result showed that MAP has the ability to slow down the speed of pomfrets' quality and inhibit the growth of bacteria.The shelf-life of pomfret could be prolonged to 3-4 days.There were mainly 6 typical bacterial strains of Pampus argenteus for physiological and biochemical identification,morphological observation,PCR Amplification and Sequencing.Gram-negative bacteria were the dominant bacteria at the beginning of storage time,but the growth of spoilage bacteria with gram-negative play a significant inhibition in MAP groups in the late periods.Shewanella putrefaciens,Pseudomonas fluorescens and Psychrobacter spp.were the most predominated spoilage bacteria of Pampus argenteus during chilled storage.Moreover,there is also a certain proportion of Brochothrix thermosphacta and Pseudomonas fragi.
