慢性应激后小鼠空间学习记忆功能及脑内BDNF表达的变化 被引量:1

摘要 目的 :研究慢性应激后小鼠海马和前脑皮层BDNF表达的变化,探讨小鼠学习记忆功能变化的机制。方法 :建立多因素慢性应激小鼠模型,通过Morris水迷宫检测小鼠空间学习记忆能力;免疫组织化学方法检测小鼠海马和前脑皮层BDNF表达水平的时序变化。结果:与对照组小鼠比较,慢性应激后即刻和24h组小鼠在水迷宫中的空间学习记忆能力明显减退(P<0.05,P<0.01),海马CA1、CA3、DG和前脑皮层BDNF的表达明显减少(P<0.05,P<0.01);而慢性应激后一周组小鼠的学习记忆能力有部分恢复,海马各区和前脑皮层BDNF的表达亦有所增加,无显著性差异。结论:BDNF在海马和前脑皮层表达的下调,可能与慢性应激后小鼠学习记忆功能的损伤有关。 Objective:To explore the mechanism of learning-memory function alteration in mice after chronic stress, and the expressions of brain derived neurotrophic factor(BDNF) of prefrontal cortex (PFC) and hippocampus (HP) in mice.Methods:The chronic stress mice model was established by using multiple chronic unpredictable stressors.The abilities of learning-memory were measured using Morris water maze task.The expression of BDNF in PFC and HP were tested by immunohistochemical method.Result:The spatial learning-memory function in mice were significantly decreased (P〈0.05, P〈0.01) in immediately and 24h after stress groups, compared with control group mice.Meanwhile,the expression of BDNF in the CA1,CA3, DG of HP and PFC were significantly reduced (P〈0.05,P〈0.01).A week after stress, the spatial learning-memory function in mice was partly enhanced, the expression of BDNF in HP and PFC were partly increased,and there was no significant difference.Conclusion:The impairment of learning-memory function induced by chronic stress in mice,which may be nearly associated with the down-regulated expression of BDNF.
