摘要
目的:探讨甲状腺癌患者血清中半乳糖血凝素-3(galectin-3)、中期因子(midkine)含量测定对肿瘤病情评估的价值。方法:收集本院收治的甲状腺癌患者108例,根据TNM肿瘤分期分为Ⅰ~Ⅱ期甲状腺癌组39例,Ⅲ期甲状腺癌组55例,Ⅳ期甲状腺癌组14例;另取同期在本院体检发现甲状腺腺瘤的患者56例作为对照组。采用酶联免疫吸附法(ELISA)测定各组血清galectin-3、midkine含量;采用荧光定量PCR测定肿瘤组织增殖基因、侵袭基因mRNA表达量;采用Pearson检验分析甲状腺癌患者血清中galectin-3、midkine含量与增殖、侵袭基因mRNA表达的相关关系。结果:甲状腺癌患者的血清glactin-3、midkine含量高于对照组,肿瘤分期越高,血清glactin-3、midkine含量越高;甲状腺癌患者肿瘤组织中增殖基因CyclinD1、BCL-2、survivin mRNA表达量高于对照组,p27mRNA表达量低于对照组,侵袭基因E-cadherin mRNA表达量低于对照组患者,S100A4、SATB1mRNA表达量高于对照组,肿瘤分期越高,上述基因mRNA表达量的变化越显著;Pearson检验显示,甲状腺癌患者血清中galectin-3、midkine含量与肿瘤组织增殖、侵袭基因表达量呈相关关系。结论:血清galectin-3、midkine含量可以直观反映甲状腺癌恶性程度,有望成为疾病早期诊断、疗效评估及预后判断的新手段。
Objective:To investigate the value of serum galectin-3 and midkine level determination for assessing tumor severity in patients with thyroid cancer.Methods:A total of 108 patients with thyroid cancer treated in our hospital were col-lected and divided intoⅠ-Ⅱ stage thyroid cancer group (n =39),Ⅲ stage thyroid cancer group (n =55)and Ⅳ stage thyroid cancer group (n =14)according to TNM stage;56 patients diagnosed with thyroid adenoma after physical examination in our hospital during the same period were selected as control group.Enzyme-linked immunosorbent assay (ELISA)was used to de-termine serum galectin-3 and midkine levels of all groups;fluorescence quantitative PCR was used to determine proliferation gene and invasion gene mRNA expression in tumor tissue;Pearson test was used to analyze the correlation of serum galectin-3 and midkine levels with proliferation and invasion gene mRNA expression in patients with thyroid cancer.Results:Serum galec-tin-3 and midkine levels of patients with thyroid cancer were higher than those of control group,and the higher the tumor stage,the higher the serum galectin-3 and midkine levels;proliferation genes CyclinD1,BCL-2 and survivin mRNA expression in tumor tissue of patients with thyroid cancer were higher than those of control group while p27 mRNA expression was lower than that of control group,invasion gene E-cadherin mRNA expression was lower than that of control group while S100A4 and SATB1 mRNA expression were higher than those of control group,and the higher the tumor stage,the more significant the change of the above gene mRNA expression;Pearson test showed that serum galectin-3 and midkine levels in patients with thy-roid cancer were correlated with proliferation and invasion gene expression in tumor tissue.Conclusion:Serum galectin-3 and midkine levels can directly reflect the malignant degree of thyroid cancer,and is expected to become the new means for early disease diagnosis,curative effect evaluation and prognosis judgment.
