摘要
目的:探讨褪黑素(MT)对大鼠急性脊髓损伤后髓过氧化物酶(MPO)及中性粒细胞趋化因子(CINC)-1表达的影响。方法:108只SD大鼠分为假手术组、脊髓损伤组、褪黑素治疗组。手术组大鼠仅切除T11椎板,脊髓损伤组、褪黑素治疗组建立T11脊髓损伤模型。脊髓损伤组、褪黑素治疗组在脊髓损伤后10 min内分别腹腔注射含体积分数5%乙醇的生理盐水和100 mg/kg的褪黑素制剂。于脊髓损伤后1 h、6 h、12 h、1 d、3 d、5 d分别取脊髓组织,化学法测定MPO活力变化,HE染色法观察脊髓组织结构,RT-PCR法观察CINC-1 mRNA表达情况。结果:脊髓内CINC-1水平和MPO活性有相似的变化规律,且MPO在CINC-1表达达峰之后12 h达高峰;除1 h外各时间点各组相比较,B组最高、C组次之、A组最低,其中B组与A组、C组差异具有统计学意义(P<0.05)。结论:褪黑素可通过抑制CINC-1的表达减少中性粒细胞的浸润,从而减轻炎症反应,起到一定的神经保护作用。
Aim: To investigate the effects of melatonin( MT) on expression of myeloperoxidase( MPO) and cytokineinduced neutrophil chemoattractant( CINC)-1 in rats after acute spinal cord injury. Methods: Totally 108 SD rats were randomly allocated into 3 groups: sham operation group( group A,n = 36),spinal cord injury group( group B,n = 36),and MT treatment group( group C,n = 36). Group B and C were established SCI animal models at T11 level. Lamina of vertebra was cut without spinal cord injury in the sham operation group. The rats of group C were injected 5%( V / V) alcohol into abdominal cavity after spinal cord injury while the rats of group B were injected MT( 100 mg / kg). Fetching the spinal tissue at 1 h,6 h,12 h,1 d,3 d and 5 d after spinal cord injury. Chemical method was used to detect the expression of MPO. HE staining was performed to observe the pathological changes of the spinal cord in the 3 groups respectively. RTPCR was used to detect the expression of CINC-1 mRNA in spinal cord tissue. Results: There were similar changing patterns of contents of CINC-1 and MPO,and the peak time of MPO was 12 h later than CINC-1. Compared the three groups except 1 h,group B had the highest level and group A had the lowest level regarding to the contents of CINC-1 and MPO,and the differences among them were statistical( P〈0. 05). Conclusion: MT could inhibit the expression of CINC-1 and reduce neutrophil infiltration,thus relieve inflammation reaction and protect spinal cord from damage.
