木犀草素对骨肉瘤干细胞增殖的影响 被引量:3

摘要 目的研究木犀草素对骨肉瘤干细胞增殖的影响。方法用免疫磁珠分选法分离人骨肉瘤细胞株MG-63中的CD133+骨肉瘤干细胞。MTT法检测0、0.01、0.02、0.04 mg/m L木犀草素溶液对骨肉瘤干细胞增殖的影响。Western blot检测木犀草素对骨肉瘤干细胞Ki67蛋白以及对骨肉瘤干细胞JAK2/STAT3信号通路的影响。结果用免疫磁珠分选法,得到比率为(87.60±5.06)%的骨肉瘤干细胞。通过MTT实验发现,与对照组比较,0.01、0.02、0.04 mg/m L木犀草素能够抑制骨肉瘤干细胞的增殖(P<0.05)。Western blot实验表明,与对照组比较,木犀草素各浓度抑制骨肉瘤干细胞Ki67蛋白的表达(P<0.05)。与对照组比较,木犀草素各浓度组对骨肉瘤干细胞JAK2/STAT3信号通路中p-JAK2和p-STAT3蛋白表达具有抑制作用(P<0.05)。结论木犀草素对骨肉瘤干细胞具有抑制作用。 Objective To explore the effect of luteolin on the proliferation of osteosarcoma stem cells. Methods CD133+osteosarcoma stem cells were separated from MG63 cells by flow cytometer. MTT was used to investigate the effects of luteolin( 0,0. 01,0. 02,0. 04 mg / m L) on the proliferation of osteosarcoma stem cells. Western blot was used to detect the levels of Ki67 protein and components of JAK2 / STAT3 signal pathway in osteosarcoma stem cells induced. Results After sorting,the content of the CD133+fraction was enriched up to( 87. 60 ± 5. 06) %. MTT assay showed that,compared with the control group,luteolin( 0. 01,0. 02,0. 04 mg / m L) inhibited proliferation of CD133+osteosarcoma stem cells( p〈0. 05). Western blot also showed that luteolin significantly decreased the level of Ki67 compared with the control group( p〈0. 05). In addition,the luteolin inhibited the expression of pJAK2 and p-STAT3 in JAK2 / STAT3 signal pathway of CD133+osteosarcoma stem cells compared with the control group( p〈0. 05).Conclusion Luteolin might be a suppressor of osteosarcoma stem cells.
