摘要
目的探讨姜黄素对体外培养人结肠癌SW480细胞增殖及细胞株中β-连环蛋白(p—catenin)、前列腺素E2(PGE2)表达的影响。方法采用不同浓度的姜黄素作用于SW480细胞,噻唑蓝法(MTT)检测细胞增殖;利用酶联免疫吸附试验(ELISA)检测药物作用后肿瘤细胞PGE,蛋白水平表达变化,蛋白免疫印迹(Westernblot)检测药物处理后B—catenin蛋白水平的表达。结果姜黄素对SW480细胞的增殖具有抑制作用,且呈剂量-时间依赖性(P〈0.05),姜黄素处理后,p-catenin、PGE2蛋白表达水平降低。结论姜黄素能够抑制SW480细胞的增殖,并下调结肠癌SW480细胞中B—catenin及PGE2的表达,这可能是其抗肿瘤作用的机制之一。
Objective To study the effect of curcumin on proliferation and expression of β-catenin and prostaglandin E2 ( PGE2 ) in human colon cancer SW480 cells. Methods SW480 cells were treated with different concentrations of curcumin and the inhibitory effect of curcumin on SW480 cells was detected by MTT. The expression of PGE2 protein were analyzed by ELISA and the expression of β-catenin protein were analyzed by Western blotting. Results Curcumin inhibited the proliferation of SW480 cells in a dose- and time-dependent manner(P 〈 0.05 ). Expression of PGE2 and β-catenin protein were decreased significantly after being treated with curcumin. Conclusion Cureumin inhibits the proliferation of SW480 cells and down-regulates of PGE2 and β-catenin protein expression,which may be one of the mechanisms of anti-cancer for curcumin.
