江西省猪细环病毒1型和2型流行情况调查及全基因组克隆与序列分析 被引量:4

Molecular epidemiology of TTSuVs in Jiangxi province and sequence analysis of full-length genomes

摘要 目前在我省许多地区猪群中是否存在TTSuV感染尚未见相关报道,为更好的了解猪细环病毒1型和2型在猪群中进化情况以及进一步研究其遗传变异,根据NCBI已发表的猪细环病毒两种基因型全基因组序列,在序列保守区域用设计特异性引物,采用聚合酶链反应(PCR)进行该病毒的检测及全基因的扩增。通过将扩增产物胶回收后进行T-A克隆,然后进行序列测定和同源性与系统进化分析。本试验检测出猪群TTSuV1型和TTSuV2型在猪群中的感染情况,全基因测序得到的序列确定为TTSuV1和TTSuV2全基因序列。对测得序列进行同源性分析与遗传进化分析,分析结果表明,分离株(JX TTSuV)与国内外不同地域TTSuV全基因组序列之间差异较大,同源性为67.8%~97.2%;进化分析表明,JX TTSuV与其他株TTSuV参考株亲缘关系远近各不相同,与日本分离株(AB076001)和巴西分离株(AY823991)亲缘关系较近。 Now To monitor the evolution Torque teno sus virus two types,and the further research on its genetic variation and the function.According to the published complete genome sequence of TTSuV1and2,special primers were designed to detect the TTSuVs and amplify the full-length viral genome by polymerase chain reaction(PCR).The purified PCR product was cloned into pMD18-T vector and then sequenced.From the sequencing analysis of the complete genome sequence of JX TTSuV,phylogenetic tree was designed.The results suggest that full-length genomes of JX TTSuV strain had 67.8%to 97.2%identity,with those of reference strains from domestic and abroad and shared the highest identity with the Japan strain(AB076001)and Brazli strain(AY823991).
