大黄素联合骨髓间充质干细胞治疗大鼠急性肝衰竭 被引量:7

Combined therapy of emodin and bone marrow mesenchymal stem cells transplantation for rat model of acute liver failure

摘要 目的探讨大黄素联合骨髓间充质干细胞对急性肝衰竭大鼠的治疗作用。方法选择SD大鼠51只,随机分4组:健康对照组6只,急性肝衰竭(ALF)模型组15只,骨髓间充质干细胞(BMSCs)组15只,大黄素联合骨髓间充质干细胞(EMD+BMSCs)组15只。D-氨基半乳糖(D-Gal N)诱导大鼠急性肝衰竭模型;BMSCs组及EMD+BMSCs组于造模后12 h尾静脉注射BMSCs悬液1.5 m L(约含BMSCs1.0×106 cells/m L),共1次;EMD+BMSCs组于造模后12 h开始腹腔注射大黄素10 mg/kg,每天2次,共3 d。ALF模型组于造模后12 h尾静脉注射等体积无菌PBS。各组大鼠均在造模后72 h,以10%的水合氯醛腹腔注射麻醉,取门静脉血及肝组织标本。测血清ALT、AST、TBil,免疫组化测肝组织Bcl-2、Bax及PCNA蛋白表达。结果造模后72 h,ALF模型组大鼠血清ALT、AST和TBil指标显著升高,与健康对照组相比,差异有统计学意义(t=7.191、6.796、6.835,P均<0.05)。与ALF模型组比较,BMSCs组及EMD+BMSCs组血清ALT、AST、TBil均有不同程度下降,以EMD+BMSCs组为著。EMD+BMSCs组中Bcl-2、PCNA表达明显高于健康对照组及ALF模型组,而Bax表达则显著低于ALF模型组,差异有统计学意义(t=5.557,P<0.05)。结论大黄素联合骨髓间充质干细胞治疗对急性肝衰竭大鼠肝功能的改善及促进肝细胞再生作用更佳,此种作用可能部分与Bcl-2/Bax的调节有关。 Objective To evaluate synergic effects of emodin combined with bone marrow mesenchymal stem cells transplantation in liver injury following acute liver failure(ALF) in rats. Methods 51 SD rats were divided into four groups. There were six rats in normal group and forty five in model group of ALF. The model of ALF (including ALF group, BMSCs group, EMD+BMSCs group, with each of 15 rats) was induced in rats by D-galactosamine (D-GaiN) successfully. Rats in BMSCs group & EMD+BMSCs group were treated with injection of 1.5 mL of suspension liquid of BMSCs (including BMSCs 1.0xl06/mL) via caudal vein 12 h after model establishment. The sera and hepatic tissue samples were collected at 72 h after D-GaiN injection. The sera were used for detecting ALT,AST and TBil. Bcl-2 and Bax in hepatic tissue samples were detected by S-P immunohistochemical staining. Results The levels of ALT,AST and TBil in model group of ALF at 72 h were significantly higher than those in normal group, and the levels of ALT,AST and TBil in every treated group were significantly reduced, especially in group EMD+BMSCs. The levels of Bcl-2 and PCNA in group EMD+BMSCs were significantly higher than those in normal and model group. The levels of Bax in group EMD+BMSCs were lower than model group. Conclusion EMD combined BMSCs are more effective to protect rats against acute liver failure,which is partly by means of down-regulating Bax expression and up-regulating Bcl-2 ex- pression in liver tissue.
