摘要
高原低氧适应性研究对于了解高原土著动物的遗传进化和高原疾病的治疗有着重要意义。HIF(hypoxia inducible factor)是氧信号转导系统的重要转录因子,在细胞的低氧应答中起着关键作用。实验旨在研究HIF-1α和HIF-2α基因表达特征在藏绵羊低氧适应中的意义。采用RT-q PCR技术对藏绵羊HIF-1a和HIF-2a基因进行了组织表达分析。结果表明:HIF-1α和HIF-2α基因在藏绵羊的8种组织(心、肝、脾、肺、肾、大脑、小脑和脑干)中均有表达。HIF-1α基因在肺和小脑中有高水平表达,其他依次是肾、脑干、大脑、心、脾和肝。HIF-2α基因在肺中表达水平最高,其他依次是脑干、心、肾、小脑、大脑和肝。结论:该研究首次证实,HIF-1α和HIF-2α基因在藏绵羊脾、大脑、小脑和脑干中均有表达,且HIF-1α在小脑中的表达水平较高。HIF-1α和HIF-2α基因在藏绵羊上呈表达广泛性和组织差异性,与其他高原土著动物有着类似的表达特征。这种个别组织HIF-1α和HIF-2α表达水平的提高提示,其正是维持低氧环境下生物体功能活动正常运转的分子基础。
Hypoxia adaptation research has important significance on understanding genetic evolution of highland animal and treatment of high altitude disease. HIF(hypoxia inducible factor) is an important transcription factor in oxygen signal transduction system, which plays a key role in cellular hypoxia response. In this study,Tibetan sheep were selected to study the expression and significance of HIF-1α and HIF-2α genes using RT-q PCR technique. The results showed that both HIF-1α and HIF-2α genes expressed in the eight tissues(heart,liver,spleen,lung, kidney,brain,cerebellum and brain stem). HIF-1α gene had high level expression in lung and cerebellum, the expression order in other tissues was kidney brain stem brain heart spleen liver. HIF-2α expression showed that the highest level was in the lung,followed by brain stem, heart, kidney,cerebellum, brain and liver. It was demonstrated for the first time that these two genes were expressed in spleen,brain,cerebellum and brain stem,and the expression level of HIF-1α in the cerebellum was relatively high in Tibetan sheep. The expression of HIF-1α and HIF-2α genes in the Tibetan sheep were extensive and with organizational differences, and had similar expression profiles with other highland indigenous animals. The increasement of expression levels of the HIF-1α and HIF-2α in some individual organizations suggested that this was molecular mechanism for maintaining normal functions of the organisms under a low oxygen environment.
