摘要
为了探讨S100A11基因在苏博美利奴羊中不同类型细度的细毛羊中的表达模式,试验以苏博美利奴羊为研究对象,克隆了钙囊素S100A11基因,采用生物信息学方法进行遗传进化分析,对S100A11蛋白的理化性质进行分析,并对其二级蛋白结构进行预测,且通过RT-PCR相对定量方法检测S100A11基因在不同绵羊个体中的表达规律。结果表明:绵羊S100A11基因的CDS全长序列为866bp,编码了105个氨基酸,与绵羊、山羊、牛、猪、鼠、兔、鸡、狗和斑马鱼的CDS同源性分别为96%、96%、96%、87%、24%、44%、70%、70%、60%;分别在1bp、220~336bp、415~864bp有3个开放式阅读框(ORF);S100A11基因全长为4798bp。S100A11蛋白含有1个信号肽、19个磷酸化位点;同时S100A11基因在超细型细毛羊皮肤组织表达量极显著高于细型细毛羊的表达量(P〈0.01),差异倍数为2.048。
To investigate the expression patterns of S100A1 gene in different types of fine- wool sheep from Subo Merino sheep,the S100A11( calgizzarin) gene was cloned using Subo Merino sheep as research object. The S100A11 gene was used for the analysis of genetic evolution using bioinformatic methods,and then the physicochemical characteristics of S100A11 protein was also analyzed. The secondary structure of S100A11 protein was predicted,and then the expression law of S100A11 gene in different sheep individuals was detected by relative quantitative RT- PCR method. The results showed that the full- length CDS sequence of S100A11 gene in Subo Merino sheep was 866 bp,encoding for a protein of 105 amino acids. The CDS of S100A11 gene shared 96%,96%,96%,87%,24%,44%,70%,70%,and 60% homologies with Ovis aries,Capra hircus,Bos taurus,Sus scrofa,Mus musculus,rabbits,Gallus domesticus,Canis lupus familiaris and Danio rerio,respectively. There were three open reading frames( ORFs) at 1 bp,220 to 336 bp,and 415 to 864 bp sites,respectively. The full- length S100A11 gene was 4 798 bp,and S100A11 protein contained 1 signal peptide and 19 phosphorylation sites. The expression level of S100A11 gene in skin tissue from superfine wool sheep were extremely significantly higher( P < 0. 01) than that in fine Merino Sheep with 2. 048- fold difference.
