猪颈动脉粥样硬化斑块易损性与Nod样受体家族包含pyrin结构域蛋白3炎性小体的关系 被引量:3

Association of NLR family, pyrin domain containing 3 inflammasome with vulnerable atherosclerotic plaques in a carotid swine model

摘要 目的 观察小型猪颈动脉粥样硬化模型中斑块组织Nod样受体家族包含pyrin结构域蛋白3(NLRP3)炎性小体的表达.方法 高脂高胆固醇饮食联合部分结扎颈动脉方法,在6头小猪建立双侧颈动脉粥样硬化模型.3个月后观察颈动脉粥样硬化斑块中NLRP3蛋白的表达.结果 12根小猪颈动脉片段中可形成不同程度的粥样硬化改变.早期斑块中,NLRP3主要表达在新生内膜的内皮细胞;进展期斑块中,NLRP3主要分布在斑块的内皮细胞、斑块基底部和脂质核心周围.NLRP3的阳性表达评分与斑块破裂[(6.65±4.32)分比(1.77±3.15)分,P<O.01]、斑块内出血[(6.18±4.31)分比(1.83±3.30)分,P<0.05]和血管新生[(6.10±4.56)分比(3.69±3.57)分,P <0.05]密切相关.结论 小型猪动脉粥样硬化斑块中NLRP3蛋白的高表达反映了斑块易损性特征. Objective To evaluate the association of NLR family,pyrin domain containing 3 (NLRP3) inflammasome expression with plaque vulnerability in a carotid atherosclerotic swine model.Methods Carotid atherosclerosis models were created in 6 Chinese miniswines using the combination of partial ligation and high cholesterol diet for 3 months.Animals were sacrificed to obtain bilateral carotid arteries for histological analysis.The relationship of morphological carotid plaque features and NLRP3 expression was also evaluated.Results Atherosclerotic plaques were developed in all 12 carotid arteries.Typical features of unstable plaque were also found.NLRP3 was expressed in both the early atherosclerotic lesions and advanced plaques.Increased NLRP3 expression was associated with the presence of plaque rupture (6.65 ±4.32 vs.1.77 ±3.15,P 〈0.01),intraplaque hemorrhage (6.18 ±4.31 vs.1.83 ±3.30,P 〈 0.05) and marked neovascularization (6.10 ±4.56 vs.3.69 ± 3.57,P 〈0.05).Conclusion Increased expression of NLRP3 may be associated with vulnerable carotid atherosclerotic plaques in a swine model.
