摘要
目的分析川乌炮制前后化学成分的变化,为深入阐明其炮制机理提供科学依据。方法采用高效液相色谱-质谱联用技术(HPLC-MS),Thermo SCIENTIFIC高效液相色谱仪,Agilent ExtendC18色谱柱,检测波长240 nm,流动相为乙腈和0.2%氨水,梯度洗脱,质谱在正离子模式下进行检测。结果生川乌中检测到99个成分,制川乌中检测到106个成分,炮制前后共有成分53个,炮制后新增成分53个,消失成分46个。川乌生制品中共鉴定成分64个。结论川乌炮制后有多种成分发生改变,本方法较高效液相色谱(HPLC)法检测6种生物碱含量能更全面地反映川乌炮制前后物质基础的变化。
Objective To contrast the components in Radix Aconiti( Common Monkshood Mother Root,Chuanwutou) before and after processing,so as to provide primary evidence for interpreting the processing principles of Radix Aconiti. Methods HPLC analysis was performed on Thermo SCIENTIFIC high performance liquid chromatograph,with Agilent Extend-C 18 column eluted with acetonitrile and 0. 2%ammonia in a gradient program at wavelength 240 nm,and mass spectrum( MS) method was conducted in the positive ion mode. Results Total 99 components were detected in unprocessed Radix Aconiti,and106 components were included in Radix Aconiti Praeparata. There were 53 in common. After processing,46 components disappeared with 53 more were detected. And 64 components were identified in unprocessed Radix Aconiti. Conclusion The change of components of Radix Aconiti before and after processing was proved by the method of HPLC-MS,which was more effective and comprehensive than the method of HPLC detecting six alkaloids.
