芪黄煎剂对大鼠胃切除术后免疫相关性肠淋巴细胞归巢数量的影响 被引量:6

Effects of Qi Huang Decoction on the number of immune correlation intestinal lymphocyte hominglympyocytes homing process of gut mucosa after gastrectomy in rats

摘要 目的:通过观察大鼠胃切除术后小肠内滴注芪黄煎剂对免疫相关性肠淋巴细胞归巢数量的影响,探讨肠淋巴细胞归巢过程及相关肠保护免疫机制。方法:选取清洁型雄性SD大鼠60只,建立模型后,将大鼠随机分为三组:对照组:大鼠胃切除手术创伤模型建立后,单纯滴注肠内营养;芪黄煎剂组:滴注的营养制剂、热量、方法、疗程均同对照组,同时滴注中药芪黄煎剂;假手术组:仅给予腹部正中切开后缝合,术后自由饮水进食。疗程结束后,除去感染死亡的模型,从假手术组、对照组、芪黄煎剂组中随机抽取15只,分别观察大鼠标本肠淋巴液中T淋巴细胞、PP结Ig A+B细胞。结果:(1)淋巴细胞归巢T细胞数量:1)CD3+T细胞:对照组低于假手术组,差异具有明显显著性(P<0.01);芪黄煎剂组同假手术组相比,差异具有显著性(P<0.05);芪黄煎剂组同对照组相比,差异具有明显显著性(P<0.01)。2)CD4+T细胞:对照组低于假手术组,差异具有明显显著性(P<0.01);芪黄煎剂组同假手术组相比,差异具有显著性(P<0.05);芪黄煎剂组同对照组相比,未见明显差异,无统计学意义(P>0.05)。3)CD8+T细胞:对照组低于假手术组,差异具有显著性(P<0.05);芪黄煎剂组同假手术组及对照组相比,均未见明显差异,无统计学意义(P>0.05)。(2)淋巴细胞归巢B细胞数量:对照组同假手术组相比较,差异具有显著性(P<0.01);芪黄煎剂组同假手术组比较,比较差异有统计学意义(P<0.05);芪黄煎剂组高于对照组,比较有显著差异(P<0.01)。结论:芪黄煎剂可以增加淋巴细胞归巢T细胞及B细胞数量,促进完成胃切除术后肠淋巴细胞归巢过程,从而保护肠黏膜免疫屏障。 Objective: To observed the influence of Qi Huang decoction on the number of intestinal lymphocyte ot gut mucosa after gastrectomy, to investigate the effect of intestinal lymphocyte homing process and protective immune mechanism. Methods: These rats were randomly divided into three groups: (1) control group: After establishment the gastrectomy trauma model, placed them in metabolic cages, they can free activities in the cages after waking, fasted on the day of surgery. Starting at the first day after operation, giving them enteral nutrition by microsyringe pump. We choosed nutrison as enteral nutrition. Dripped of saline 6ml before dripping nutrient solution.(2) Qi Huang decoction group: intravenous injection of nutritional formulations, heat, methods and treatments was the same as the simply en- teral nutrition group, meanwhile dripping Qi Huang decoction. (3) sham group: Only suturing after abdominal midline incision, without gastrectomy. Free water to eat after the operation, without giving enteral nutrition and Chinese tradi- tional medicine.Before infusion of the nutrient solution can be given vegan Qi Huang Decoction 6ml, injected into the small intestine uniformly and continuously for a week. Results (1)Lymphocytes homing T cell number(CD3 + T cells and their subsets CD4 +, CD8 + T cells changes in rats lymph : After modeling rats a week): after 1 week, the drainage of lymph cells by the Flow Cytometer.(1)CD3 + T cells: the control group were lower than control group, has obvious significant difference (P〈O.01). compared with control group, with significant difference (P〈0.O5);Qi Huang decoction group compared with control group, has obvious significant difference (P〈0.O1).2) the CD4+T cells: the control group were lower than control group, has obvious significant difference (P〈0.01). Qi Huang decoction group compared with control group, has no significant difference, no statistical significance (P〉O.05). (3) CD8 + T cells: control group were lower than control group, with significant difference (P〈O.05). compared with control group and the control group, no obvious difference, has no statistical significance (P〉0.05). (2) Lymphocytes homing B cells (The number change of IgA + B ceils in PP junction of the small intestine of rats): the control group compared with control group, with significant difference (P〈O.01); compared to control group, compare the difference was statistically significant (P〈0.05); Qi Huang decoction group was higher than control group, the more significant differences (P〈O.01). Conclusion: Traditional Chi- nese medicine Qi Huang decoction can increase the number of lymphocyte homing T ceils and B ceils, promote the in- testinal lymphocyte homing process, protect the intestinal mueosal immune barrier.
