摘要
目的探讨莱菔硫烷(SFN)对阿尔茨海默病(AD)大鼠海马超微结构和白介素(IL)-1β水平的影响。方法 42只Wistar大鼠随机分成对照组,模型组,治疗组(吡拉西坦430 mg/kg),SFN高、中、低剂量组(SFN 40,20,10 mg/kg)。采用Aβ1~42双侧脑室注射大鼠诱导建立AD模型。给予SFN连续灌胃治疗35 d后取材,观察大鼠海马镜下超微结构、计算脾和胸腺脏器指数,采用双抗体夹心法测定海马组织IL-1β水平。结果与模型组比较,SFN高、中剂量组脑海马形态明显改善,脾和胸腺脏器指数明显增加(P〈0.05),海马组织IL-1β水平明显降低(P〈0.05)。结论SFN能改善AD大鼠海马形态结构,增加脾和胸腺脏器指数,降低海马组织IL-1β水平。
Objective To observe the effects of sulforaphane( SFN) on ultrastructure and IL-1β of hippocampal in Alzheimer's disease( AD) rat. Methods Wistar rats were randomly divided into control,model,treatment( Piracetam 430 mg / kg),SFN high-dose( 40 mg/kg),middle-dose( 20 mg/kg),low-dose( 10 mg/kg) groups. Aβ1 ~ 42 was injected into the bilateral hippocampal of rat to induce AD models. The rats were killed after 35 days continuous treating. After the treatment,all animals were sacrificed,ultrastructure of hippocampal was observed,calculated the organ coefficients of splenic and thymus. Biochemical methods were used to test the content of IL-1β. Results Compared with that of model group,the brain maintain normal morphological structure of hippocampal in SFN high-dose and medial-dose groups was significantly increased,the organ coefficients of splenic and thymus significantly were increased( P 0. 05),the level of IL-1β of hippocampal tissue were significantly decreased( P 0. 05). Conclusions SFN could maintain normal morphological structure of hippocampal of AD rat,increase organ coefficients of splenic and thymus,decrease IL-1β responses in the hippocampal tissue.
