摘要
了解带叶兜兰菌根真菌的群落组成,为优良菌株的筛选奠定基础,对采自广西乐业县的野生带叶兜兰进行了菌根真菌分离,通过根段直接培养法分离菌根真菌,以确定75%乙醇和10%NaClO的最适表面消毒时间,并应用形态学方法对分离的菌株进行了属的鉴定。研究结果显示,带叶兜兰菌根真菌最适表面消毒时间是75%乙醇8 min,10%NaClO 8 min。从带叶兜兰菌根中分离到的63株真菌,除8株菌株分类地位不能确定外,其余菌株归属于镰刀菌属(Fusarium)、轮层菌属(Daldinia,)、碳团菌属(Hypoxylon)、毛壳菌属(Chaetomiaceae)、胶膜菌属(Tulasnella)5属,其中镰刀菌属和轮层菌属为优势菌属。
In order to understand the community composition of myeorrhizal fungi in Paphiopedilum hlrsutissimun(Orchidaceac) and establish foundation for screening excellent strains, the optimal method for sterilization of plant materials surface with 75 % ethanol and 10 % NaClO was determined by the tissue isolation method and fungus was identified preliminarily based on morphological characteristics in this paper. The results showed that 75 % ethanol for 8 rain, 10 % NaCIO for 8 rain were more suitable for roots sterilization. 63 strains fungi were isola- ted except that 8 strsains remained undetermined, other 55 strains were classified and belonged to 5 genera. The dominant groups of those mycorrhizal fungi were Fusarium and Daldinia, and Hypoxylon were firstly reported on Paphiopedilum hirsutissimun. Specifically, Tulasnel- la, a species of typical orchid myeorrhizal fungl,has been found.
