褪黑素通过抑制P-选择素表达减轻内毒素致大鼠急性肺损伤 被引量:3

Melatonin inhibited P-selectin expression on acute lung injury induced by lipopolysaccharide in rats

摘要 为探讨P-选择素(Ps)在内毒素(LPS)致大鼠急性肺损伤(ALI)时肺组织中的表达及褪黑素(MT)对其表达的影响,将SD大鼠随机分为3组,对照(Control)组、模型(LPS)组、褪黑素干预(MT+LPS)组,采用气管内滴注脂多糖(LPS)的方法建立大鼠ALI模型,光镜下观察大鼠肺组织形态学改变,采用免疫组织化学染色技术、逆转录-聚合酶链反应(RT-PCR)和Western blot技术观察肺组织中P-选择素的表达变化.结果显示:模型组较对照组肺组织损伤严重,P-选择素的阳性表达明显增多(P<0.05);应用褪黑素干预后显著缓解上述变化(P<0.05).这说明褪黑素可能通过抑制P-选择素的表达发挥对急性肺损伤的保护作用. To observe the expression of P-selectin(Ps) in lung tissues of acute lung injury(ALI) rat model induced by lipopolysaccharide(LPS) and examine the effect of melatonin(MT) to its expression all rats were randomly divided into three groups:Control group, LPS group and MT+LPS group. Lung tissues were imbedded by paraffin to observe morphological changes. The auther used immunohistochemical staining technique, reverse transcription- polymerase chain reaction ( RT - PCR ) and western blot method to detect the expression of P-selectin. The results showed that compared with Control group,alveolar septum thickened significantly in LPS group,positive expression of P-selectin was very obvious(P〈0.05) ;the ad- ministration of MT mitigated above changed significantly(P〈0.05). The results suggested that MT pos- sessed obvious protective effect on lung tissues during ALI,its protective mechanism might be related to the inhibition of the expression of P-selectin.
