摘要
目的:研究巨细胞病毒(CMV)宫内感染对子代豚鼠海马神经元Tau蛋白磷酸化的影响。方法:建立豚鼠CMV宫内感染动物模型,PCR法筛选CMV DNA阴性豚鼠,雌雄同笼受孕;孕龄1-20d时,随机分成两组:感染组腹腔内接种CMV病毒悬液,对照组接种生理盐水,足月分娩的子代豚鼠喂养至3月龄后无菌采集脑组织,应用免疫组织化学法和免疫印迹法检测各组子代豚鼠海马神经元Tau、p-Tau(Ser 404)、PKA及PP2A蛋白表达水平。结果:与对照组相比,感染组子代豚鼠海马神经元Tau、p-Tau(Ser 404)及PKA蛋白的表达水平明显上升,而PP2A蛋白的表达水平明显下降,差异具有统计学意义(P<0.01)。结论:CMV宫内感染导致子代豚鼠海马神经元Tau蛋白磷酸化水平增加,可能与PKA及PP2A蛋白的表达异常相关。
Objective: To study Tau protein phosphorylation of the descendant guinea pig with cytomeg- alovirus (CMV) intrauterine infection. Methods: The model of CMV intrauterine infection was built as follows: The female and male guinea pigs without CMV-infected identified by PCR were chosen and mated for pregnancy. The pregnant were randomly divided into two groups in gesta- tional age from one to twenty: CMV was injected into abdominal cavity in CMV-infected group, and saline was injected in control group. These full term offspring were fed for 3 months to col- lect brain tissue in sterile conditions. The protein expression of Tau,p-Tau(Ser 404),PKA, and PP2A were measured by immunohistochemistry and Western blotting. Results: Compared with those in control group, the protein expressions of Tau,p-Tau(Ser 404) ,and PKA in CMV-infec- ted group significantly increased, while the protein expression of PP2A significantly decreased (P 〈0.01). Conclusion: CMV intrauterine infection can increase Tau protein phosphorylation, and the abnormal expression of both PKA and PP2A might be involved in the process.
