摘要
解淀粉芽孢杆菌CGMCC-9021能够高效降解黄曲霉毒素B_1(Aflatoxin B_1,AFB_1),确定其降解AFB_1的活性蛋白及其机理是今后工业化生产应用的重要基础。通过硫酸铵分级沉淀、DEAE FF阴离子交换层析和Superdux 75凝胶过滤层析对解淀粉芽孢杆菌CGMCC-9021发酵液中降解AFB_1的活性物质进行分离纯化,通过Tricine-SDS-PAGE和质谱鉴定初步分析该活性蛋白为裂解酶(分子质量约27 ku)。同时,采用荧光色谱技术初步分析了裂解酶对AFB_1的降解机理可能是破坏AFB_1的内酯环结构。
Bacillus amyloliquefaciens CGMCC-9021 can biodegrade the AFB1 efficiently. Study on its active protein and possible mechanism is prerequisite to industry application. The active protein was purified from fermented liquid of B. amyloliquefaciens CGMCC-9021 by ammonium sulfate precipitation,followed by DEAE FF and Superdux 75 gel filtration chromatography. The purified protein was identified as lytic enzyme( Mr 27 ku) by Tricine-SDS-PAGE and tandem mass spectrometry( MS / MS). In addition,the fluorescence spectrometry of AFB1 degradation product was studied. Results showed the possible mechanism of AFB1 biodegradation was that the lactone ring structure of AFB1 was cleaved by the lytic enzyme.
