摘要
目的研究不同粒径、不同材料、不同形状的纳米粒子对蛋白质分子结构的影响。方法通过原子力显微镜表征纳米粒子吸附蛋白质前后的粒径变化,研究其对蛋白质结构的影响。结果从粒径大小来看,纳米粒子的粒径越小对蛋白质的结构造成的影响越小;从纳米粒子的材料来看,半导体材料的纳米粒子对蛋白质结构造成的影响大于金纳米粒子;从纳米材料的形状来看,球形纳米颗粒对蛋白质结构造成的影响小于片状石墨烯。结论纳米粒子的粒径、材料和形状对蛋白质的分子结构有着显著的影响。
Objective To study the effects of different characteristics of nanoparticles on the structure of protein. Methods Using atomic force microscopy (AFM) to characterize the changes of nanoparticles sizes before and after protein adsorption, and then the effects on the structure of protein were determined. Results In the aspect of the nanoparticles size, the binding of protein onto smaller nanoparticles resulted in smaller change of the protein structure. In the aspect of the materials of nanoparticles, effect of the semiconductor nanoparticles on the structure of protein was greater than that of the gold nanoparticles. In the aspect of the shape of nanomaterial, the nanosheets of graphene oxide (GO) had greater influence on the deformation of protein structure than spherical nanoparticles. Conclusion The particle size, material, and shape have significant effects on the structure of protein.
