HPLC测定不同产地玛咖中苄基芥子油苷的含量 被引量:6

Determination of benzyl glucosinolate in Lepidium meyenii from different regions by HPLC

摘要 分析不同产地玛咖中苄基芥子油苷的含量,为玛咖的质量标准提供依据。利用葡糖硫苷酶酶解苄基芥子油苷,采用HPLC测定酶解产物异硫氰酸苄酯的含量,色谱条件为Kaseisorb LC ODS 2000色谱柱(4.6 mm×150 mm,5μm),流动相为乙腈(A)-水(B),以1.0 m L·min-1的流速进行梯度洗脱,0~5 min,3%~8%A,5~9 min,8%~48%A,9~23 min,48%~62%A,23~28 min,62%~99%A,进样量为10μL,检测波长246 nm,柱温40℃。不同产地玛咖中苄基芥子油苷质量分数介于10.76~17.91 g·L-1。该方法简便、准确,重复性好,适用于玛咖中苄基芥子油苷的含量测定。 The content of benzyl isothiocyanate( BITC) which as the enzymatic hydrolysis product of benzyl glucosinolate through thioglucosidase was determined by HPLC. The content of benzyl isothiocyanate( BITC) which as the enzymatic hydrolysis product of benzyl glucosinolate through thioglucosidase was determined by HPLC. The chromatography condition was as follows: Kaseisorb LC ODS 2000( 4. 6 mm × 150 mm,5 μm) column with the mobile phase of acetonitrile( A)-water( B) under gradient elution( 0-5 min,3%-8% A; 5-9 min,8%-48% A; 9-23 min,48%-62% A; 23-28 min,62%-99% A); the flow rate was 1. 0 m L·min- 1with 10 μL injection volume; detection wavelength was 246 nm and temperature of column was 40 ℃. The content of benzyl glucosinolate was in the range of 10. 76-17. 91 g·L^(-1). The method is simple,accurate and good reproducibility which can be used for the determination of benzyl glucosinolate in Lepidium meyenii,effectively.
