摘要
为了探讨内皮祖细胞(EPCs)移植对睾丸扭转(TT)复位后患侧睾丸的影响,获取大鼠骨髓源性EPCs,进行分组:对照组(sham组,左侧睾丸不扭转)、缺血再灌注损伤组(IRI组,左侧TT 720°2 h后复位。)、EPCs移植组(EPCs组,睾丸复位后经股静脉注射EPCs悬液)。术后5 d,左侧睾丸称重,冰冻切片技术检测EPCs归巢,HE染色检测左侧睾丸组织病理变化,Makler评分(TMS)评价睾丸生精功能。术后5 d,EPCs归巢于左侧睾丸。IRI组、EPCs组左侧睾丸重量低于sham组(P<0.05),EPCs组左侧睾丸重量高于IRI组(P<0.05)。IRI组左侧睾丸病理损伤严重,EPCs组左侧睾丸病理损伤较IRI组减轻。IRI组、EPCs组TMS均低于sham组(P<0.05),EPCs组TMS高于IRI组(P<0.05)。TT可致患侧睾丸生精功能受损,EPCs移植可改善患侧睾丸生精功能障碍。
In order to investigate the effect of endothelial progenitor cells (EPCs) transplantation on function of ipsilateral testes after testicular torsion, bone - marrow - derived EPCs were obtained from rats. The rats were di- vided into 3 groups: control group (sham group, the left testes were not twisted), ischemia reperfusion injury group(IRI group, they were in detorsion after the left testes were twisted 720° for 2 h), endothelial progenitor cells transplantation group( EPCs group, EPCs suspension was injected into rats through the femoral vein after testicular detorsion). On the 5th day after operation, the rats'left testes were weighed, EPCs homing was detec- ted by using the technique of frozen section. The pathological changes in the left testes were examined by HE staining technique. Spermatogenesis was evaluated by testicular Makler score(TMS). On the 5th day after opera- tion, transplanted EPCs homed into the left testes. The left testicular weight in rats of the IRI group and EPCs group was significantly lower than that in the sham group ( P 〈 0.05 ). The left testicular weight in the EPCs group was higher than that in the IRI group, the difference was significant ( P 〈 0.05 ). The left testis underwent serious pathological changes in IRI group. Compared with IRI group, the impaired changes in the left testes from the EPCs group mitigated. TMS in the IRI group and EPCs group was significantly lower than that in the sham group(P 〈 0.05). TMS in rats of EPCs group was higher than that in the IRI group(P 〈 0.05 ). TT can impair spermatogenesis in twisted testis. EPCs transplantation is effective for improving the spermatogenic disfunction in twisted testes.
