TGFβ1在大黄多糖治疗糖尿病皮肤创面中的作用 被引量:3

The role of TGFβ1 in rheum polysaccharide-treated diabetic wound surface

摘要 目的探讨大黄多糖对糖尿病皮肤创面的疗效及可能机制。方法链脲佐菌素诱导糖尿病大鼠模型,2周后手术制作背部创面模型,再随机分为糖尿病组(给予生理盐水)、大黄多糖组(给予大黄多糖)及大黄多糖-SB431542组(给予大黄多糖及TGFβ1受体抑制剂SB431542混合液),另取正常大鼠制作背部创面模型给予生理盐水为对照组。2周后观测创面愈合率,检测创面羟脯氨酸含量,HE染色观测创面肉芽组织生长情况,免疫组化染色检测TGFβ1表达。结果与对照组相比,糖尿病组及大黄多糖-SB431542组创面愈合率明显降低,羟脯氨酸含量减少(均<0.01),而大黄多糖组无明显变化(>0.05)。对照组与大黄多糖组真皮层较厚,胶原纤维丰富,而糖尿病组与大黄多糖-SB431542组真皮层较薄,皮下胶原组织较少。糖尿病组TGFβ1表达较对照组减少,大黄多糖组及大黄多糖-SB431542组TGFβ1表达明显增加。结论诱导TGFβ1表达是大黄多糖促进糖尿病皮肤创面愈合的重要机制之一。 Objective To explore the role and possible mechanism of rheum polysaccharide(RP) in the recovery of diabetic wound surface of rat. Methods The diabetic rats were induced by streptozotocin, and wound surface in back was performed 2 weeks later. The animals were randomly assigned into diabetic group(N.S.), RP group(RP), RP+SB431542 group(RP+TGFβ1 receptor inhibitor SB431542), the na?ve rats with back wound surface administered with N.S. were considered as control group. The healing ratio, contents of hydroxyproline(OHP), granulation tissue and the expressions of TGFβ1 were revealed by HE and immunohistochemistry respectively. Results Compared with control group, the healing ratio and content of OHP were decreased in diabetic group and RP-SB431542 group(P〈 0.01, respectively), with no alteration in RP group(P〉 0.05). The thicken dermis and abundant collagen fibers were observed in control and RP groups, while thin dermis and little collagen fibers were detected in diabetic and RP-SB431542 groups. The expression level of TGFβ1 was lower in diabetic group than in control group, but higher in RP and RP-SB431542 groups. Conclusion TGFβ1 contributes to the therapeutic effect of RP on diabetic wound surface.
