摘要
利用响应面分析方法研究铁皮石斛总多酚的最佳提取工艺及体外抗氧化性。在选取酒精浓度、料液比、温度做单因素实验的基础上,进行三因素三水平的Box-Behnken中心组合研究,建立了影响因素与总多酚之间的函数关系。获得最佳工艺条件为:乙醇浓度75%,料液比1∶40,提取温度75℃;在此最佳条件下水浴条件下浸提1 h,总多酚含量为28.01 mg/g,实验结果与模型预测值相符。铁皮石斛多酚提取物具有较强的清除对DPPH·和·OH自由基能力,其IC50(半抑制浓度)分别为526.872μg/m L和1263.024μg/m L。
The optimum extraction conditions of total polyphenols from Dendrobium officinale by response surface methodology,as well as their antioxidant activities in vitro,were studied in the research. The experiment method was designed according to Box-Behnken central composite design with 3 factors and 3 levels on the basis of single-factor experiment of ethanol concentration,material-to-liquid ratio and temperature. The relationship between influencing factors and response values was subsequently established. The results indicated the optimum extraction conditions were as follows:the extracting solvent was 75% ethanol,material-to-liquid ratio was 1∶40,extracting temperature was 75 ℃,and extracting time was 1 h. Under these conditions,the maximal yield of total polyphenols reached 28.01 mg/g,which was well matched with the predicted content. Moreover,the polyphenols extracted from Dendrobium officinale exhibited an effective scavenging effect on DPPH· and·OH free radicals, and its IC50( half inhibitory concentration) was respectively up to 526. 872 μg / m L and1263.024 μg/m L.
